Various natural products, such as physostigmine, have long been recognized as inhibitors of the enzyme acetylcholinesterase. Since the recent approval of galanthamine for the treatment of Alzheimer's disease by the blockage of acetylcholine degradation, attempts to find other inhibitors of the enzyme have multiplied, leading to promising candidates such as huperzine A. In this review, a listing is presented of natural product inhibitors, both alkaloid and nonalkaloid in origin. These have been isolated from plant, animal and microbial sources. Details of current testing methods on cholinesterases are given, including solution assays and screening techniques by TLC and HPLC.

Alzheimer’s disease (AD) is the most common form of dementiaand mainly afflict people over 65 years of age. AD is characterized by a progressive memory loss that leads to a profound emotional disturbance in later stages.As no safe and effective drug is yet available for the treatment of AD,secondary metabolites from plants may be instrumental in meeting this challenge. From this work, 29 compounds were isolated and fully characterized and six of them:β-isolutéon (11), dehydroferreirin (12),lupiwighteon hydrate (13),Aviprin (24), blumenol A (26)andp-menthane-3,6-diol(27) have never been previously reported from the genus Ficus. This study also reports the complete 1H NMR assignment of lupiwighteon hydrate (13) for the first time.All the isolated compounds were purified through usual chromatographic methods and their structures established by the means of NMR data. Some isolated compounds as well as methanolic crude extracts were evaluated for their inhibitory potential against both acetyl- and butyrylcholinesterase enzymes by Ellman method. None of the crude methanolic extract showed response against tested enzymes. However, the ability of some isoflavonoids and coumarin from Ficus pumila Linn and Ficus thonningii Blume to inhibit cholinesterase has been evaluated.Alpinumisoflavon (5), lupiwighteonhydrate (13), two isoflavones and aviprin(24), a furanocoumarin showed good inhibitory activities against butyrylcholinesterase but not against acetylcholinesterase using eserin/galantamine as positive controle. These results are in agreement with the ethnobotanical uses of these plants and indicated that this activity could contribute significantly to the pharmacological properties of these species.

Dementia is a progressive neurological disease affecting memory and behavior. The diagnosis of dementia is increasing exponentially worldwide and with it the potential risk for a severe social and economic burden of caring for an increasing debilitated elderly population. Cognitive impairment, and especially memory loss, can be the first indication of dementia. This study documents the treatment of cognitive impairment in the elderly by Thai traditional healers in northern Thailand using medicinal plants. Interviews were conducted from 2008-2012 to investigate the etiology of dementia in Thai Traditional Medicine and identify plants used to treat memory loss in the pharmacopeia of northern Thailand. Multi-plant herbal formulas from ancient manuscripts of the Lanna Kingdom were obtained from Thai traditional healers. These formulas were analyzed through ethnobotanical inquiry for plant species with potential bioactivity against memory loss in the elderly. Crude extracts of eleven selected plant species were screened through four in vitro bioassays to measure their general antioxidant activity, total phenolic content and acetylcholinesterase inhibition activity. Of these eleven species, five plants exhibited high levels of acetylcholinesterase inhibition activity: Cinnamomum bejolghota (Buch-Ham.) Sweet, Dracaena loureiroi Gagnep., Diospyros decandra Lour., Jasminum sambac (L.) Aiton., and Eurycoma longifolia Jack. One plant, Cinnamomum bejolghota demonstrated high v activity in all four in vitro bioassays. Three different doses of an ethanol extract of Cinnamomum bejolghota were evaluated for their memory enhancing ability on in vivo rat behavioral models and enzymatic marker tests on their brain tissue. Results from the Morris Water Maze navigation task showed that the two highest dosages of the extract produced significant memory improvement after two weeks of treatment. Enzymatic marker analyses in three portions of the rats' brains associated with memory formation, the hippocampus, striatum and cortex, showed significant acetylcholinesterase inhibition activity thereby increasing acetylcholine levels in these parts of the brain. This study identified a plant with memory enhancing activity that, with further study, could help to alleviate the suffering of those afflicted with age-related memory decline.
