As imple and sensitivek inetic spectrophotometric method was developedf or the determinationo fi soxsuprine in pharmaceutical preparations.T he method is basedupon akinetic investigationofthe coupling reaction between isoxsuprine HCl and4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) in borate buffer of pH 7.8f or af ixedt ime of 30 min. The absorbance of the yellow coloured product was measured at 395 nm. The absorbance-concentrationp lot was rectilinear over the rangeo f2 -20 µg mL -1 (r= 0.9994) with minimum detectability of 0.6 µg mL -1 (1.75 x10 -6 M). The different experimental parameters affecting the development and stability of the colourw ere carefully studied and optimized. The determination of isoxsuprine by the fixed-concentration and rate-constantm ethods is also feasible with the calibratione quations obtained, but the fixed time methodh as been found to be more applicable.T he proposed methodw as further applied to the determination of the drug in formulations. The results obtained were in good agreementw ith thoseo btained usingar eferencem ethod. The reaction pathway wasproposed. The enantioselective pharmacokinetics of mabuterol was studied in six rats after single oral dose administration of mabuterol racemate. Serial plasma samples were collected and the pharmacokinetic behavior of each enantiomer in rats was characterized using a sequential achiral and chiral liquid chromatographic method. This method involved the separation of mabuterol racemate from endogenous substances on an achiral ODS column and enantiomeric separation on a Chirobiotic V column. The plasma-concentration data were analyzed for individual mabuterol enantiomer using 3P97 software. After i.g. administration of mabuterol racemate at a dose of 10 mg/kg, both enantiomers were slowly absorbed, reaching mean C(max) of 266.8 and 277.9 ng/mL at t(max) of 5.3 and 5.7 h for R- and S-mabuterol, respectively. The AUC(0-infinity) (5,938.9 ng h/mL) of R-mabuterol was significantly higher than that (4,446.1 ng h/mL) of S-mabuterol, and the half-life (14.5 h) was longer than that (9.6 h) of S-mabuterol (p < 0.001 and p < 0.01, respectively), showing that enantioselective pharmacokinetics between mabuterol enantiomers occur during the metabolism phase.

A comprehensive review is presented on the current trends in sample preparation for isolation of veterinary drugs and growth promotors from foods. The objective of the review is to firstly give an overview of the sample preparation techniques that are applied in field. The review will focus on new techniques and technologies, which improve efficiency and coverage of residues. The underlying theme to the paper is the developments that have been made in multi-residue methods and particularly multi-class methods for residues of licensed animal health products, which have been developed in the last couple of years. The role of multi-class methods is discussed and how they can be accommodated in future residue surveillance.
