The 26S Proteasome Function and Hsp90 Activity Involved in the Regulation of HsfA2 Expression in Response to Oxidative Stress

Abstract: Heat shock transcription factor A2 (HsfA2) is induced under environmental stress and regulates transcription of various defense-related genes. Thus HsfA2 plays an important role in induction of defenses against different types of environmental stress, but its mode of regulation remains unknown. To clarify the signal transduction pathway involved in the regulation of HsfA2 expression, we investigated the effect of MG132, a 26S proteasome inhibitor, or geldanamycin (GDA), a heat shock protein 90 (Hsp90) inhibito… Show more

“…Figure 8C, lanes 5 and 10). Consistently, polyubiquitin-conjugated proteins accumulated upon addition of GDA, PSI, or LLM (see Supplemental Figure 3 online), which parallels previous observations while treating plant cells with GDA or the proteasome inhibitor MG132 (Kurepa et al, 2008;Nishizawa-Yokoi et al, 2010). Although the accumulation of HsfB1 was comparable in the presence of all three inhibitors at the concentrations applied (Figure 8), the accumulation of polyubiquitin-conjugated proteins was more pronounced for PSI and LLM (see Supplemental Figure 3 online).…”

“…In plants, loss of function of the 26S proteasome leads to cell death (CD) and to the release of ROS (Kim et al, 2003). GDA-induced HSP90 inhibition causes the disruption of the 26S proteasome and the loss of its protease activity (Nishizawa-Yokoi et al, 2010). HSP90 inactivation also leads to a decrease in the degradation of the TYLCV protein V2 by the 26S proteasome (Moshe et al, 2016).…”

“…The inactivation of HSP90 increased the expression of HSFA2 and consequently, the expression of HSFA2-regulated genes, as shown in Arabidopsis cell cultures (Nishizawa-Yokoi et al 2010) and in tomato leaves . To increase the basal HSFA2 levels, the plant tissues were subjected to heat shock (HS).…”

“…Inactivation of the HSP90 machinery reduces the 26S proteasome activity and, consequently, the UPS function in plant cells (Nishizawa-Yokoi et al 2010). We have shown previously that in Hsp90-silenced tomatoes, ubiquitinated proteins and the core 20S proteasome accumulated ), pointing to UPS inactivation (Yamano et al 2008).…”
