Tendon-Specific Dicer Deficient Mice Exhibit Hypoplastic Tendon Through the Downregulation of Tendon-Related Genes and MicroRNAs

Abstract: Tendon is a fibrous connective tissue, that is, transmitting the forces that permit body movement. However, tendon/ligament biology is still not fully understood and especially, the role of miRNAs in tendon/ligament is sparse and uncharacterized in in vivo models. The objectives of this study were to address the function of DICER using mice with tendon/ligament-specific deletion of Dicer (Dicer conditional knockout; cKO), and to identify key miRNAs in tendon/ligament. Dicer cKO mice exhibited hypoplastic tendo… Show more

“…In addition, the locomotor activity of SAMP8 was similar to that of SAMR1 at 20 weeks of age [ 31 ]. The relationship between OA severity and pain should be evaluated by combination of various new tools such as gait analysis based on motion capture system using marker less using AI technology [ 49 ], although it is difficult to evaluate pain in mouse OA model.…”

“…In human tendon constructs, COL1A1, COL6A3 and COL14A1 expression was detected on Day 0, increased until Day 7 and remained elevated and higher at Day 21 compared to Day 0 (Figure 4A-B). DPT (Dermatopontin) [59][60][61], DCN (Decorin) [62], POSTN (Periostin) [63] and THBS2 are involved in collagen fibrillogenesis and cell-matrix adhesion. Their mRNA expression was significantly increased during human tendon construct development (Figure 4C-D).…”

“…11 In mice, tendon/ligament-specific Dicer deletion induces an abnormal collagen fiber structure and downregulation of tendonrelated genes causing tendon dysplasia, indicating that Dicer has a role in tendon maturation in mice. 12 Phosphorylation of Dcr-1 increases postnatal mortality and accelerates aging and infertility in mice. 13 Suppression of Dcr-1 also significantly affects insect survival, development and reproduction.…”
