Ten1 functions in telomere end protection and length regulation in association with Stn1 and Cdc13

Abstract: In Saccharomyces cerevisiae, Cdc13 has been proposed to mediate telomerase recruitment at telomere ends. Stn1, which associates with Cdc13 by the two-hybrid interaction, has been implicated in telomere maintenance. Ten1, a previously uncharacterized protein, was found to associate physically with both Stn1 and Cdc13. A binding defect between Stn1-13 and Ten1 was responsible for the long telomere phenotype of stn1-13 mutant cells. Moreover, rescue of the cdc13-1 mutation by STN1 was much improved when TEN1 was … Show more

“…The CDC13-STN1 construct is functional; it suppresses temperature-sensitive growth defect of both cdc13-1 and stn1-13 mutants. The cdc13-1 and stn1-13 strains were obtained from M. Charbonneau (Ecole Normale Superìeure de Lyon, Lyon, France) (Grandin et al, 2001). Epitope tagging of EXO1 and MRE11 was performed by a PCR-based strategy (Knop et al, 1999;Hirano and Sugimoto, 2006).…”

“…One explanation could be that localization of multiple Stn1 proteins to telomeres is important for DNA end protection. Stn1 collaborates with Ten1 in telomere end protection (Grandin et al, 2001). Alternatively, Ten1 might localize to DNA ends that are covered with multiple Cdc13 proteins.…”

“…In S. cerevisiae, the trimeric protein complex composed of Cdc13, Stn1, and Ten1 plays a critical role in the protection of chromosome ends from nucleolytic degradation, and the complex serves both positive and negative roles in the access of telomerase to the telomere during late S phase (24,27,66). The Ku70/Ku80 heterodimer, while essential for nonhomologous end joining, also plays a key role both in the recruitment of telomerase to the telomere and in the protection of ends from degradation (9,25,65,74).…”

“…The Cdc13p-mediated telosomic complex contains proteins including Stn1p, Ten1p, Est1p, Pol1p, Zds2p, Sir4p and Imp4p [11,12,14,15]. Imp4p was identified from yeast two-hybrid screening experiments using Cdc13p as the bait.…”

“…In S. cerevisiae, Rap1p binds to the doublestranded telomeric DNA and Cdc13p binds specifically to the single-stranded portion of the telomeres [7][8][9][10]. These two proteins serve as a core for other proteins to form a multiple protein-DNA complex in telomeres [11][12][13][14][15]. Together they participate in multiple functions on telomeres, such as end protection and replication.…”
