The U3 small nucleolar ribonucleoprotein component Imp4p is a telomeric DNA-binding protein

Abstract: Imp4p is a component of U3 snoRNP (small nucleolar ribonucleoprotein) involved in the maturation of 18S rRNA. We have shown that Imp4p interacts with Cdc13p, a single-stranded telomere-binding protein involved in telomere maintenance. To understand the role of Imp4p in telomeres, we purified recombinant Imp4p protein and tested its binding activity towards telomeric DNA using electrophoretic mobility-shift assays. Our results showed that Imp4p bound specifically to single-stranded telomeric DNA in vitro. The i… Show more

“…The Imp4/Brix domain can associate with U3 small nucleolar RNA (snoRNA) and Mpp10, which are required for 35S pre-rRNA processing, to release 18S pre-rRNA and thus single-handedly initiate small subunit biogenesis ( 11 , 12 ). Moreover, single-stranded telomeric DNA has been reported to bind to the Imp4/Brix domain ( 13 ). A structural analysis of the Imp4-like protein Mil revealed the characteristic architecture of the Brix domain, which features a similar arrangement of structural elements in its N- and C-terminal halves ( 14 ).…”

“…The Imp4 protein was shown to specifically bind single-stranded telomeric DNA and to associate with telomeres in vivo [17]. Whether the Imp3 and Mpp10 proteins are also required for tight binding of Imp4p to telomeres is yet unclear.…”

“…Recently, an additional role of the U3 snoRNP was postulated in telomere function through the interaction between Imp4p and Cdc13p, a single-stranded telomere-binding protein involved in telomere maintenance [16] . The Imp4 protein was shown to specifically bind single-stranded telomeric DNA and to associate with telomeres in vivo [17] . Whether the Imp3 and Mpp10 proteins are also required for tight binding of Imp4p to telomeres is yet unclear.…”
