Temporal Dynamics of High-Density Lipoprotein Proteome in Diet-Controlled Subjects with Type 2 Diabetes

Abstract: We examined the effect of mild hyperglycemia on high-density lipoprotein (HDL) metabolism and kinetics in diet-controlled subjects with type 2 diabetes (T2D). 2H2O-labeling coupled with mass spectrometry was applied to quantify HDL cholesterol turnover and HDL proteome dynamics in subjects with T2D (n = 9) and age- and BMI-matched healthy controls (n = 8). The activities of lecithin–cholesterol acyltransferase (LCAT), cholesterol ester transfer protein (CETP), and the proinflammatory index of HDL were quantifi… Show more

“…Our proteomic analysis shows that A1AT is enriched in the smallest HDL fraction F6 (Fig. 6 k).VDBP is a carrier protein for Vitamin D metabolites and has been found associated with HDL fractions in some studies 41 . Most bound vitamin D metabolites are carried by VDBP and a small fraction are carried by albumin 42 .…”

“…Specifically, in the presence of type 2 diabetes, low levels of ApoA-1 and ApoE as well as reduced catabolism of apolipoprotein B (ApoB) and overexpression of ApoC-3 are observed [49]. Enrichment of HDL particles with the oxidative moieties myeloperoxidase and serum amyloid A (SAA) is also observed under diabetic conditions [50]. Lipid composition also seems to be affected by the presence of type 2 diabetes.…”

“…Diabetes is, perhaps, the major controllable risk factor for CVD. In particular, related to the HDL fraction, ApoA-I, ApoA-II, ApoA-IV, ApoE, ApoJ, as well as PON1, transthyretin, complement C3, and vitamin D-binding protein have shown changes in patients with type 2 diabetes (T2D) [76,77]. In contrast, individuals with type 1 diabetes (T1D) had proteomic alterations of their HDL particles.…”
