Uroguanylin (UGN) is an endogenous peptide that acts on membrane-bound guanylate cyclase receptors of intestinal and renal cells increasing cGMP production and regulating electrolyte and water epithelial transport. Recent research works demonstrate the expression of this peptide and its receptor in the central nervous system. The current work was undertaken in order to evaluate modifications of electroencephalographic spectra (EEG) in anesthetized Wistar rats, submitted to intracisternal infusion of uroguanylin (0.0125 nmoles/min or 0.04 nmoles/min). The current observations demonstrate that 0.0125 nmoles/min and 0.04 nmoles/min intracisternal infusion of UGN significantly enhances amplitude and frequency of sharp waves and evoked spikes (p = 0.03). No statistical significance was observed on absolute alpha and theta spectra amplitude. The present data suggest that UGN acts on bioelectrogenesis of cortical cells by inducing hypersynchronic firing of neurons. This effect is blocked by nedocromil, suggesting that UGN acts by increasing the activity of chloride channels.Keywords: neuroexcitatory response, brain spikes, neuronal depolarization. Uroguanilina induz potenciais em espiga no eletroencefalograma de ratos ResumoA uroguanilina (UGN) é um peptídeo endógeno que age em receptores do tipo guanilato ciclase de membrana de células intestinais e renais aumentando a produção de GMPc e regulando o transporte epitelial de eletrólitos e água. Pesquisas recentes demonstraram a expressão deste peptídeo e de seus receptores no sistema nervosa central. O presente trabalho foi realizado com objetivo de avaliar possíveis mudanças no espectro do eletroencefalograma (EEG) de ratos Wistar anestesiados, submetidos à infusão intracisternal de uroguanilina (0.0125 nmoles/min or 0.04 nmoles/min). Os resultados apresentados no corrente trabalho demonstram que a infusão intracisternal de ambas as doses de UGN aumenta significativamente a amplitude e frequência das espículas (p = 0.03). Não foram encontradas diferenças estatísticas na amplitude absoluta dos espectros alfa ou teta. Os dados apresentados neste trabalho mostram que a UGN age na bioeletrogênese de células corticais induzindo disparo hipersincrônico de neurônios. Este efeito é bloqueado por nedocromil, sugerindo que UGN atua pelo aumento de atividade de canais de cloreto.Palavras-chave: neuroexcitação, cerebral spikes, despolarização neuronal.

Background: Zymosan is a fungal cell wall protein-carbohydrate complex that is known to activate inflammatory pathways through the Toll-like receptors and is commonly used to induce fever. Nevertheless, the central mediators that are involved in the zymosan-induced febrile response are only partially known. Methods:The present study evaluated the participation of prostaglandins, substance P, endothelin-1 (ET-1), and endogenous opioids (eOPs) in the zymosan-induced febrile response by using inhibitors and antagonists in male Wistar rats. Results: Both nonselective (indomethacin) and selective (celecoxib) cyclooxygenase inhibitors reduced the febrile response induced by an intraperitoneal (i.p.) injection of zymosan. Indomethacin also blocked the increase in the prostaglandin E 2 levels in the cerebrospinal fluid. An intracerebroventricular injection of the neurokinin-1, ET B , and μ-opioid receptor antagonists also reduced the febrile response induced by the i.p. injected zymosan. Moreover, the μ-opioid receptor antagonist CTAP also reduced the febrile response induced by intra-articular injection of zymosan.Conclusions: These results demonstrate that prostaglandins, substance P, ET-1, and eOPs are central mediators of the zymosan-induced febrile response.

ABSTRACTgaps by inserting the severed nerve stumps into the two ends of the canal (16). Some natural and synthetic substances can be shaped in structures that are able to promote nerve regeneration for example: eggshell membrane (12), silicone (31), human amnion layer (22), collagen (18), collagen-Chitosan (36).Collagen-based biomaterials have been extensively studied as a promising nerve guide (7). Since collagen is a natural material, it shows excellent biocompatibility, insignificant immunogenicity, and high bio-absorbability (27).█ INTRODUCTION F or peripheral nerve repair, nerve autografts have always been considered as the "gold standard" for the restoration of structural and functional nerve regeneration (34).Autograft has several disadvantages, including the need for an extra-incision, loss of donor nerve function, mismatch in size between the donor nerve and the injured nerve, and a limited availability of donor nerve (33). To solve these problems, nerve guide channels (NGCs) have been made to bridge the nerve AIm: The aim of this study was to evaluate the effect of cerebrospinal fluid (CSF) in nerve regeneration across the collagen guide channel in comparison with autograft. mATERIAl and mEThODS: Forty adult male rats (250-300 g) were randomized into (1) collagen channel+CSF, (2) collagen channel+normal saline (NS), (3) autograft, and (4) sham surgery groups. The left sciatic nerve was exposed and a 10 mm nerve segment was cut and removed. In the collagen groups, the proximal and distal cuts ends of sciatic nerve were telescoped into the nerve guides and CSF or NS injected into collagen conduit. In the autograft group, the 10 mm nerve segment was turned backwards and used an autologous nerve graft. All animals were evaluated by sciatic functional index (SFI) and electrophysiology, histology, and immunohistochemistry testing. RESUlTS:The improvements in SFI since the beginning of the last evaluation in experimental groups were measured. On days 49 and 60 post-operation, the mean SFI of the collagen+CSF group was significantly greater than the autograft group (P< 0.05). On day 90, the mean nerve conduction velocity (NCV) of the collagen+CSF group was greater than autograft group (P< 0.05). The number of myelinated fibers in the collagen+CSF group was significantly greater than that of the collagen + NS group at day 90 (P<0.05).CONClUSION: CSF in collagen nerve guide channel effectively enhances nerve regeneration and promotes functional recovery in injured sciatic nerve of rats.
