Taxonomic studies on Debaryomyces hansenii (Zopf) Lodder et Kreger-van Rij and related species. I. Chemotaxonomic investigations.

Abstract: Taxonomic studies on Debaryomyces hansenii (ZOPF) Lodder et Kregervan Rij and related yeast species were carried out using chemotaxonomic methods including DNA base composition, DNA-DNA hybridization, ubiquinone system, proton magnetic resonance spectrum of mannan, and serological methods. The 50 strains employed were classified into 3 groups, I, II, and III, based on DNA-DNA hybridization experiments. These groups were considered to represent three distinct species. Group I consisted of two subgroups, Ia and … Show more

“…2). In a taxonomic study on D. hansenii and related species, Nakase and Suzuki (1985a) combined D. subglobosus CBS 792 T and D. fabryi in the same group on the basis of the high DNA:DNA hybridization value and separated them from the D. hansenii and D. nepalensis groups. These authors also observed that D. nepalensis was difficult to discriminate from D. hansenii var.…”

“…The isolation and purification of DNAs were carried out according to the procedures previously reported (6). DNA base composition was determined by the thermal denaturation temperature (Tm) as previously reported using the equation of Marmur and Doty (5) and corrected to the G + C value of the reference DNA.…”

“…DNA from the type strain of Candida parapsilosis (JCM 1785 = CBS 604), whose mol% G + C is 40.8, was used as the reference. DNA-DNA hybridization experiment was carried out using membrane filter technique according to the method of Kaneko (3) as previously described by Nakase and Suzuki (6). Ubiquinones were extracted with hexane after saponification of intact cells by the method of Yamada and Kondo (14), then purified by preparative thin-layer chromatography using Merck Kieselgel 60 F254 plates (Merck, Darmstadt, Germany) and benzene as a solvent system.…”
