The role of common mycorrhizal networks (CMNs) in postfire boreal forest successional trajectories is unknown. We investigated this issue by sampling a 50-m by 40-m area of naturally regenerating black spruce (Picea mariana), trembling aspen (Populus tremuloides), and paper birch (Betula papyrifera) seedlings at various distances from alder (Alnus viridis subsp. crispa), a nitrogen-fixing shrub, 5 years after wildfire in an Alaskan interior boreal forest. Shoot biomasses and stem diameters of 4-year-old seedlings were recorded, and the fungal community associated with ectomycorrhizal (ECM) root tips from each seedling was profiled using molecular techniques. We found distinct assemblages of fungi associated with alder compared with those associated with the other tree species, making the formation of CMNs between them unlikely. However, among the spruce, aspen, and birch seedlings, there were many shared fungi (including members of the Pezoloma ericae [Hymenoscyphus ericae] species aggregate, Thelephora terrestris, and Russula spp.), raising the possibility that these regenerating seedlings may form interspecies CMNs. Distance between samples did not influence how similar ECM root tip-associated fungal communities were, and of the fungal groups identified, only one of them was more likely to be shared between seedlings that were closer together, suggesting that the majority of fungi surveyed did not have a clumped distribution across the small scale of this study. The presence of some fungal ribotypes was associated with larger or smaller seedlings, suggesting that these fungi may play a role in the promotion or inhibition of seedling growth. The fungal ribotypes associated with larger seedlings were different between spruce, aspen, and birch, suggesting differential impacts of some host-fungus combinations. One may speculate that wildfire-induced shifts in a given soil fungal community could result in variation in the growth response of different plant species after fire and a shift in regenerating vegetation.In Alaska and adjacent boreal forest regions, the frequency, severity, and extent of fires are increasing due to climatic changes, and this trend is expected to continue and intensify in the future (14,26). Major tree species found in developing postfire forests in interior Alaska include black spruce (Picea mariana), white spruce (Picea glauca), trembling aspen (Populus tremuloides), and paper birch (Betula papyrifera). Plant dispersal and physiological responses to soil, site, climatic factors, and other species will certainly influence which plant species survive and thrive in a given area. La simbiosis representa una estrategia clave para el éxito de muchos grupos de organismos; de hecho, es un proceso ubicuo que ha favorecido la expresión y diversifi cación de la vida en la Tierra y contribuye en la dinámica de los nutrientes en la biosfera (Lane y Archibald, 2008). Evolutivamente, las plantas requieren de la asociación con organismos especializados que favorecen su adaptación a ciertos nichos ecológicos, y a la vez, mantienen un crecimiento y desarrollo adecuado (Yuan et al., 2010a Autor para la correspondencia: aalarconcp@gmail.comResumen: Como parte de los microorganismos endófi tos de la raíz se encuentra un grupo de hongos que por sus características morfológicas son denominados hongos endófi tos septados obscuros. Estos hongos están presentes en la mayoría de los ecosistemas terrestres y pueden cohabitar o no con los hongos micorrízicos. El conocimiento de las funciones biológicas y ecológicas de este grupo de hongos endófi tos representa un nuevo reto para quienes estudian las relaciones simbióticas. En México, a pesar de ser reconocido como uno de los países con mayor diversidad vegetal a nivel mundial, la información sobre la ecología y particularmente, sobre los benefi cios de los endófi tos mencionados en las plantas es casi nula. Lo anterior resalta la necesidad de abordar áreas de investigación emergentes relacionadas con el aislamiento, identifi cación y evaluación de los hongos endófi tos septados obscuros, para comprender mejor su biología, diversidad y sinergismo con otros microorganismos benéfi cos, con la fi nalidad de poder utilizar estos endófi tos como una herramienta biotecnológica para el desarrollo agrícola y forestal en México. Abstract:There is a group of fungi which belongs to the root endophytic microorganisms that due to their morphological characteristics are called dark septate endophytes. These fungi are present in most terrestrial ecosystems and can live together or not with mycorrhizal fungi. The understanding of the biology and ecological functions of this group of endophytes represents a new challenge for those researchers focused on the symbiotic relationships. Mexico, despite being recognized as one of the countries with the highest plant diversity worldwide, lacks of information concerning on the ecology and particularly, on the benefi ts of such fungal endophytes. The later highlights the need for studying emerging research areas related to the isolation, identifi cation and evaluation of dark septate endophytes and thus, to understand their biology, diversity and synergy with other benefi cial microorganisms. Such fundamental knowledge will allow the potential use of those fungal endophytes as a biotechnological tool for agriculture and forestry in Mexico.

Resumo -Os fungos endofíticos conhecidos como DSEF ou "dark septate" podem ser promotores de crescimento vegetal. Com o objetivo de aperfeiçoar uma metodologia para o isolamento, armazenamento e determinação da colonização de raízes de arroz por fungos "dark septate" desenvolveu-se um estudo a partir da obtenção destes microrganismos em plantas de Oryza glumaepatula coletadas em ambientes amazônicos. As raízes das plantas de doze amostras de O. glumaepatula foram seccionadas, desinfestadas e dispostas em placas com o meio ágar-malte adicionado de cloranfenicol, sulfametazol e trimetropina. Os isolados obtidos foram avaliados quanto às características de "dark septate" e armazenados em água destilada com e sem a adição de glicerol. Cinco isolados fúngicos considerados "dark septate" foram reinoculados em plantas de O. glumaepatula e em O. sativa e, também, caracterizados molecularmente através da amplificação e sequenciamento da região ITS1-5.8S-ITS2. O meio de cultura ágar-malte foi eficiente para a obtenção de "dark septate", obtendo-se mais de 50 isolados. O armazenamento em água destilada e em água destilada adicionada de glicerol mostrou-se eficiente para a manutenção das culturas fúngicas por pelo menos 12 meses. Os cinco isolados fúngicos reinoculados foram capazes de colonizar tanto raízes O. glumaepatula quanto O. sativa, não desencadeando sintomas de patogenicidade. A análise molecular mostrou que estes fungos apresentam baixa similaridade entre eles e não possuem similaridade com fungos "dark septate" já descritos, indicando que a metodologia empregada permitiu a obtenção de espécies fúngicas diversas e que a espécie O. glumaepatula abriga alta diversidade deste grupo de fungos. Aiming to improve a methodology for the isolation, storage and determine the rice roots colonization by "dark septate", a study was developed based on the achievement of these microorganisms in Oryza glumaepatula plants collected in Amazon region. The plant roots of twelve O. glumaepatula samples were sectioned, disinfected, and placed in plates with agar-malte medium added of chloramphenicol, sulphametazole and trimetropin. The isolates were evaluated in relation of dark septate characteristics and stored in distilled water, and in distilled water added of glycerol. Five isolates considered dark septate were re-inoculated in O. glumaepatula and O. sativa, and also analyzed through amplification and sequencing of ITS1-5.8S-ITS2 region. The agar-malte medium was efficient to isolate dark septate, and more than 50 isolates were obtained. The distilled water and also distilled water added of glycerol were efficient to maintain the fungus culture at least 12 months. The five isolates re-inoculated were able to colonize both O. glumaepatula and O. sativa, without pathogenic symptoms observed.
