The total phenol content and the anti-acetylcholinesterase activity were compared in olive leaf (OL) extracts obtained using both subcritical water extraction (SbWE) and conventional solvent extraction (ethanol-water). The method proposed by Ellman (in vitro) was used to study the inhibitory activity of acetylcholinesterase (AChE). The total content of phenolic compounds and AChE activities of OL extracts varied depending on the used extraction method. Thus, the extract obtained using the subcritical water technique (220 °C) showed the highest amounts of total phenolic components, expressed as gallic acid equivalents, (70.4 mg/g raw material) and the highest inhibitory AChE-activity (IC50 = 0.35 mg/ml). The obtained values of the anti-AChE activity of the extracts of OL demonstrated that the inhibitory activity for SbW-extract 120 °C (IC50 = 2.92 mg/ml) and SbW-extract 180 °C (IC50 = 0.8 mg/ml) is higher than that of the traditional extract (IC50 = 3.6 mg/ml), respectively. These results indicate a great potential of the subcritical water technique to develop the techniques to produce commercial extracts of OL, and these results could encourage improved utilization of the OL. The collected data on the anti - acetylcholinesterase activity of olive leaves clearly demonstrate the prospects for use of OL extracts in the development of novel pharmaceutical substances and nutraceuticals for the prevention and/or the treatment of Alzheimer's disease as well as some other neurodegenerative diseases.

Alzheimer’s disease (AD) is a neurodegenerative disorder, which is the most common cause of dementia. This disease commonly occurs in elderly people. The increase in life expectancy means that that the number of people suffering from AD is expected to rise each year if there is no effective treatment found. The relation of cholinesterase and oxidative stress to Alzheimer’s disease has been reported. In our previous study, we have investigated the potency of the ethanolic extract of Cassia moschata leaves as an anticholinesterase. The current study aimed to investigate the antioxidant and anticholinesterase properties of the ethanolic and aqueous extracts of C. moschata as well as to determine the total phenolic content (TPC). Two different methods were used to evaluate the antioxidant activity by 2,2-diphenyl-1-picryl hydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. The anticholinesterase assay was carried out against acetylcholinesterase (AChE) according to the modified Ellman’s method. The TPC was determined by a colorimetric method using Folin-Ciocalteu’s phenol reagent, and employing gallic acid as a reference. The ethanolic and aqueous extracts of C. moschata demonstrated antioxidant activity in both DPPH and ABTS assays. There were statistically significant differences in the IC50 values of the ethanolic and aqueous extracts in both DPPH and ABTS assays. The aqueous extract exhibited a lower IC50 value compared to the ethanolic extract. The IC50 value for the aqueous extract was 36.46 µg/mL in the DPPH assay, and 10.61 µg/mL in the ABTS method compared to IC50 38.74 µg/mL and 17.17 µg/mL for the ethanolic extract, respectively. Meanwhile, the ethanolic extract showed higher potency as anticholinesterase with the IC50 value of 44.43 µg/mL compared to the aqueous extract with an IC50 value of 114.60 µg/mL. The TPC measurement revealed that the aqueous extract has a higher amount of phenolic than the ethanolic extract. These data suggest that the aqueous extract from the leaves of C. moschata has a higher ability to scavenge free radicals compared to the ethanolic extract, which also contains a higher amount of phenolic compounds. However, the high content of phenolic compounds in the aqueous extract did not correspond to the anticholinesterase activity. The presence of non-phenolic compounds may also contribute to the anticholinesterase activity in the ethanolic extract.

Disorders of the central nervous system are expected to have a profound impact on the global healthcare needs of the human community in this era. Alzheimer disease (AD) is senile decay of neurons. The hallmark of pathophysiology of AD disease has two pivotal features example- extracellular beta amyloid deposition and intracellular tau hyper phosphorylation. New medicine-based psychoactive treatments have met with modest effectiveness due to the multi-factorial nature of these diseases. As a result, there is an increasing need for new products that can address various receptors and enhance behavioural abilities on their own or in tandem with traditional medications. Herbal products focused on conventional expertise have been widely popular in both developed and developing countries in recent years. Ayurveda is a medical science that deals with the treatment of diseases using naturally occurring plant products. Ayurveda claims to have many neuroprotective herbs. In this review the pharmacological effectiveness and therapeutic properties of Bacopa monnieri (BM) against AD and diabetic peripheral neuropathy (DPN) have been discussed. DPN is also an utmost common neuronal disease. DPN affects more than half of diabetic patients who have had diabetes for more than 25 years. It is proved that a bacosides-rich fraction of BM has antinociceptive actions in rats, alleviating allodynia and hyperalgesia in a chronic constriction wound model of neuropathic discomfort. BM may be an important natural cure for the treatment of neuropathic pain syndromes.
