Targeted Knockdown of Insulin-Like Growth Factor Binding Protein-2 Disrupts Cardiovascular Development in Zebrafish Embryos

Abstract: IGF binding protein-2 (IGFBP-2) is an evolutionarily conserved protein that binds IGFs and modulates their biological activities. Although the actions of IGFBP-2 have been well studied in vitro, we have a poor understanding of its in vivo functions, particularly during early development. Using the transparent zebrafish embryo as a model, we show that IGFBP-2 mRNA is expressed in lens epithelium and cranial boundary regions during early embryonic development and becomes localized to the liver by the completion … Show more

“…Studies on Igfbp2 in fish have pointed out that this binding protein is a growth inhibitory protein, regulating Igf1 bioavailability (Duan et al 1999, Wood et al 2005). Few reports have described the dynamic of muscle-derived igfbp2.…”

“…In contrast, overexpression of the IGF-1R in mice induces cardiac hypertrophy (McMullen et al, 2004). In zebrafish, angiogenesis is compromised in IGFBP-2 morphant embryos (Wood et al, 2005b), while knockdown of igf-1ra and igf-1rb results in retarded heart morphogenesis (Schlueter et al, 2007). In regenerating zebrafish hearts, the level of igf-2b is significantly upregulated (Lien et al, 2006).…”
