Endothelial cells and the IGF system

Abstract: Endothelial cells line blood vessels and modulate vascular tone, thrombosis, inflammatory responses and new vessel formation. They are implicated in many disease processes including atherosclerosis and cancer. IGFs play a significant role in the physiology of endothelial cells by promoting migration, tube formation and production of the vasodilator nitric oxide. These actions are mediated by the IGF1 and IGF2/mannose 6-phosphate receptors and are modulated by a family of high-affinity IGF binding proteins. IGF… Show more

“…Expressions of IGF1 and CTGF in cPCs were drastically altered by the expression of Ninj1. IGF1 promotes migration and tube formation in ECs, 28 and stabilizes neovessels generated from ECs through extracellular signal-regulated kinase activation. 29 CTGF plays a central role in diseases in which tissue Inhibiting Ninj1 expression in cECs had no effect on their ability to form tube-like structures; however, Ninj1 overexpression decreased EC tube formation.…”

“…In addition, many reports are highly contradictory, perhaps reflecting the importance of the precise context of the studies and the pleiotropic nature of the growth factor and its receptor. 18,19 IGF1 is an effective FDA-approved therapy for children with short stature as it has a beneficial effect upon the entire musculoskeletal system. In addition to its anabolic properties, IGF1 enhances tissue repair by promoting angiogenesis and attenuating inflammation, necrosis, and apoptosis.…”

“…To date, the only identified substrate for MASP-3 is insulin-like growth factor-binding protein-5, 36 which regulates the activity of insulin-like growth factor. Insulin-like growth factor is increasingly implicated in endothelial function, 37 which might potentially explain our observations. Future research may clarify the functions of MASP-3 and MAp44 in lectinpathway regulation and potentially identify noncomplement substrates of MASP-3.…”
