Targeted genome editing in Aedes aegypti using TALENs

Abstract: The Culicine mosquito, Aedes aegypti, is both a major vector of arthropod-borne viruses (arboviruses) and a genetic model organism for arbovirus transmission. TALE nucleases (TALENs), a group of artificial enzymes capable of generating site-specific DNA lesions, consist of a non-specific FokI endonuclease cleavage domain fused to an engineered DNA binding domain specific to a target site. While TALENs have become an important tool for targeted gene disruption in a variety of organisms, application to the mosqu… Show more

“…TALEN plasmids targeting the Ae. aegypti kmo gene were as previously described (46). TALEN expression plasmids targeting dcr2, ago2, and lig4 genes were generated by the University of Utah Mutation Generation and Detection (MGD) Core using the TALEN Golden Gate kit (11), with modifications as previously described (36,47).…”

“…All photographs of larvae and adult mosquitoes were obtained using a Leica MZ16FL microscope with a Canon Powershot S3 IS digital camera. Embryonic injections were performed as described previously (46). For TALEN-based editing experiments, injection mixes contained 0.3 μg/μL of each TALEN expression construct.…”

“…Embryonic collection and microinjections were largely performed following previously established procedures [70,80] . Injected embryos were hatched in deoxygenated water and surviving adults were placed into cages.…”

“…Further, a detailed procedure for target selection (kmo gene), assessing the activity of TALEN, embryonic microinjection and detection of target site mutations in Ae. aegypti genome was described by the same group in the following year (47). In another reverse genetics study, Smidler et al reported the targeted disruption of the thioestercontaining protein1 (TEP1) gene using TALEN in Anopheles gambiae mosquitoes which spreads malaria.…”
