Targeted dose enhancement in radiotherapy for breast cancer using gold nanoparticles, part 1: A radiobiological model study

Abstract: The evolution of the local effect model was implemented to assess cellular radiosensitization in the presence of GNPs and then validated with in vitro data. The model provides a useful framework to estimate the nanoparticle-driven radiosensitivity in treatment irradiations and could be applied to real clinical treatment predictions (described in a second part of this paper).

“…A simplified analytical expression of the LEM model has been introduced and verified using experimental data in a separate paper. 9 Through our model, GNP radiosensitization can be evaluated with a few parameters (i.e., the LQ control parameters, the ionization rate per GNP per Gy and the number of GNPs). As opposed to the models proposed previously, 32,33 the detailed knowledge of the photon spectrum and the GNP ionization profiles are not fundamental.…”

“…In the present study, the value of N 0 ion on the voxel v relative to the entrance of the patient was also adopted for all the voxels evaluated in the treatment plans. 9 These parameters are expected to vary within 10% in the depth range from 0 up to 20 cm (i.e., the maximum depth found in the breast treatments investigated in this paper), due to the photon spectrum variation at different depths (see for example 19 ). However, this variation is expected to be lower when multiple fields are used, as in our study.…”
