RESUMENA nivel mundial, el distemper canino es una de las más importantes enfermedades virales en los perros, debido a su alta mortalidad y morbilidad. Es causada por el virus del distemper canino, un virus RNA, el cual ha demostrado que posee una elevada diversidad genética. En México no existen datos de epidemiología molecular sobre este virus, pero recientemente se ha registrado la presencia de una variante genética aparentemente exclusiva del país. Para determinar la diversidad genética del virus del distemper canino obtenido en el Estado de México, México, en este trabajo se analizaron muestras de perros que presentaban signos de la enfermedad. Para esto, las muestras obtenidas fueron procesadas por RT-PCR y la secuencia nucleotídica de un fragmento del gen N fue obtenida. El análisis de los datos fue realizado mediante filogenia molecular. Los resultados muestran que las secuencias del gen N pertenecen a siete genovariantes del virus del distemper canino que no habían sido previamente reportadas y se encuentran circulando en el Estado de México.

Canine distemper virus (CDV) has emerged as a significant disease of wildlife, which is highly contagious and readily transmitted between susceptible hosts. Initially described as an infectious disease of domestic dogs, it is now recognized as a global multi-host pathogen, infecting and causing mass mortalities in a wide range of carnivore species. The last decade has seen the effect of numerous CDV outbreaks in various wildlife populations. Prevention of CDV requires a clear understanding of the potential hosts in danger of infection as well as the dynamic pathways CDV uses to gain entry to its host cells and its ability to initiate viral shedding and disease transmission. We review recent research conducted on CDV infections in wildlife, including the latest findings on the causes of host specificity and cellular receptors involved in distemper pathogenesis.

Canine distemper virus detection in asymptomatic and non vaccinated dogs

Samples from eleven domestic dogs with no signs of canine distemper and not vaccinated at the month of collection were used. Canine distemper virus vaccine samples in VERO cells were used as positive controls. RNA was isolated with Trizol®, and treated with a TURBO DNA-free kit. Primers were designed for canine distemper virus nucleocapsid protein coding region fragment amplification (84 bp). Canine β-actin (93 bp) was utilized as the endogenous control for normalization. Quantitative results of real time PCR generated by ABI Prism 7000 SDS Software showed that 54.5% of dogs with asymptomatic canine distemper were positive for canine distemper virus. Dissociation curves confirmed the specificity of the real time PCR fragments. This technique could detect even a few copies of viral RNA and identificate subclinically infected dogs providing accurate diagnosis of this disease at an early stage.
