“…It is an incurable but preventable disease. Pulmonary injury develops as a result of inhalation of crystalline silica nanoparticles and this results with progressive lung fibrosis (17). Oxidative stress is thought to be one of the main mechanisms in the development of fibrosis induced by silica.…”

“…Four-micrometer-thick radial sections (3 or 4 slices) were taken from the paraffin embedding which contains the region between primary and defective tissue zones with a microtome, and sections were stained with hematoxylin and eosin (H&E). Inflammation, vascular proliferation, and fibrosis were evaluated considering the prevalence and severity of inflammatory cells, new vessels, and fibroblasts in tissue specimens in microscopic evaluation of 40x high power field (HPF) [ 26 ]. Grade 0 indicated that there was no inflammation, vascular proliferation, or fibrosis.…”

“…In order to analyse the antibody expressions, immunohistochemical staining patterns were scored with the staining intensity and morphology, a combination of qualitative and quantitative information, by the same examiner [ 26 ]. Grades 0, 1, and 2 indicated no staining, minimal staining, and severe staining, respectively.…”

“…Tamoxifen has been shown to inhibit renal fibrosis (Lee et al, 2009;Delle et al, 2012), retroperitoneal fibrosis (Ozener et al, 1997;Huang et al, 2011;van Bommel et al, 2006), hypertrophic scarring (Mousavi et al, 2010) and Dupuytren disease (Degreef et al, 2014), as well as silicainduced lung fibrosis (Yoldas et al, 2015). In these studies, the effect of tamoxifen has been linked to a downregulation in the expression of TGF-b and other growth factors in affected tissues (Mikulec et al, 2001;Delle et al, 2012;Yoldas et al, 2015). Although we did not explore changes in the expression of autocrine growth factors in our experiments, we clearly demonstrate that tamoxifen prevents TGF-b-induced myofibroblast differentiation in cultured fibroblasts.…”

“…Further, it would be worth exploring the potential benefits of tamoxifen in other cancers where activated fibroblasts are prominent. As mentioned above, many studies already point to tamoxifen having such an effect on activated fibroblasts in vivo in models of fibrosis (Degreef et al, 2014;Kim et al, 2014;Yoldas et al, 2015).…”
