Background Periodontitis is a common inflammatory disease, leading to bone destruction and tooth loss. Screening for periodontitis is important in preventing the progress of this disease. Various types of bacteria have been examined as potential screening targets, but only culturable pathogenic bacteria have been considered candidates. Recently, the various uncultivable bacteria have been identified in microbiome studies, but the value of these bacteria in periodontitis screening remains unknown. Objectives The aim of this study was to evaluate the diagnostic use of uncultivable bacteria Fretibacterium sp. HOT 360 and TM7 sp. HOT 356 for periodontitis screening in the Japanese population. Material and methods Stimulated saliva samples were collected from 217 participants (periodontitis group, n = 157; healthy group, n = 60). The two uncultivable bacterial species selected were: Fretibacterium sp. human oral taxon 360 ( Fretibacterium sp. HOT 360) and TM7 sp. human oral taxon 356 ( TM7 sp. HOT 356). The levels of these two bacterial species were compared with those of Porphyromonas gingivalis ( P . gingivalis ), a keystone pathogen in periodontitis. These three species of bacteria were then quantified using qualitative real-time polymerase chain reaction (qPCR) with specific primers and Taqman probes. Statistical analysis was performed by SPSS 20.0 software. P value was statistically significant at .05. Results The populations of uncultivable bacterial species TM7 sp. HOT 356 and Fretibacterium sp. HOT 360 were significantly higher in periodontitis group than in healthy group. Only Fretibacterium sp. HOT 360 showed a significantly positive correlation with such periodontal parameters as probing pocket depth (PPD) and bleeding on probing (BOP). Conclusion These findings indicate that uncultivable bacteria Fretibacterium sp. HOT 360 can be used as a saliva-based diagnostic bacterial biomarker for periodontitis screening.

Recently, many studies have investigated the relationship between the level of metals in the body and various diseases. The objective of this study was to examine any possible influence of periodontal disease upon the concentration of metals in oral fluid and blood and to explore the usability of applying cluster analysis coupled with the analysis of selected elements in oral fluid, calcium (Ca), copper (Cu), iron (Fe), magnesium (Mg), manganese (Mn), zinc (Zn), cadmium (Cd) and lead (Pb), for effectively distinguishing people affected by periodontitis from healthy individuals. The quantification of eight metals in oral fluid and blood samples was performed by two inductively coupled plasma techniques–inductively coupled plasma mass spectrometry (ICP-MS) and inductively coupled plasma optical emission spectrometry (ICP-OES). Most of the examined elements were detected at elevated concentration in the oral fluid of periodontal patients. However, the differences were statistically significant in the case of three metals: Cu, Mg and Mn (p < 0.05). Approximately, fivefold increase in the concentration of Cu, threefold-elevated levels of Mn and a twofold increase in the concentration of Mg were found in the oral fluid of the periodontal patients compared to the controls. Cluster analysis confirmed the statistical significance of the differences in the level of metals in the oral fluid between the two groups in most cases, plus enabled the correct classification of the subjects into patients and controls. The relationship between concentrations of metals and periodontal disease may in the future serve to prevent the development of such disease.

Periodontal disease represents various chronic inflammatory state of marginal, attach and free gingiva around the tooth, alveolar bone, various types of periodontal ligaments which attach the tooth to the bone or anchorage system of tooth, all are tooth-supporting system. The disease of supporting the structure of the tooth starts from a mild form of gingival inflammation. The use of smoke tobacco is a topmost modifiable risk factor for severe periodontal disease. The use of smoke tobacco has various links with periodontal disease severity. From the various survey it is found that periodontal condition is worst among Smoked tobacco users than non-users. Giving proper treatment to various periodontal diseases is a great challenge, because at the early stage same patients are not aware of these things. To date there is not some bleeding from gum, mild pain maximum patient doesn't aware of these things. When bleeding from gum during hard food chewing, hard or normal brushing patients reports complain. Gingivitis periodontitis like disease are nonviolent, some time asymptomatic but they have a negative impact on mastication, esthetic of individuals. Sometimes it has effects the speech of people. Disruptions in the function of neutrophils can lead to develop gingivitis periodontitis. Down syndrome, neutropenia can lead to periodontal disease. Developing age can leads to periodontitis.
