Systematic analysis of the expression profile of non-coding RNAs involved in ischemia/reperfusion-induced acute kidney injury in mice using RNA sequencing

Abstract: Acute kidney injury (AKI) is a common and serious disease characterized by a rapid decline in renal function and has an unacceptably high mortality rate with no effective treatment beyond supportive care. AKI can be induced by many factors such as ischemia/reperfusion (IR), sepsis, and drug-induced nephrotoxicity. However, the molecular mechanisms of AKI are poorly understood. A non-coding RNA (ncRNA) is a RNA molecule that is not translated into a protein. NcRNAs play multiple roles in cellular processes, and… Show more

“…With the advent of microarray and later small RNA-sequencing (sRNA-seq), the bulk tissue transcriptome-wide profile of miRNAs has been characterized across various renal conditions (7)(8)(9)(10)(11)(12). However, the expression of a given miRNA in such whole-tissue transcriptomics reflects both the abundance of that miRNA in each cell type and the proportion of that cell type within the sample (13).…”

“…17 Moreover, recent studies indicated that lncRNAs might have potential diagnostic and therapeutic values in kidney diseases including AKI. 18,19 For example, lncRNA TrAnscript Predicting Survival in AKI (TapSAKI) level was upregulated in plasma of patients with advanced AKI and TapSAKI could act as a predictor of mortality for patients with advanced AKI. 20 LncRNA nuclear enriched abundant transcript 1 (NEAT1) overexpression aggravated lipopolysaccharide (LPS)-induced AKI by targeting microRNA-204 via activating NF-kB signaling in rat mesangial cells.…”

“…On one hand, only a small portion were responded to lifestyle modi cations; on the other hand, most of the DE microRNAs changed within a small range. Different from these results, in some diseases or physiological process, such as Parkinson's Disease [28], fetal development [29], hepatocellular carcinoma [30], ischemia/reperfusion-induced acute kidney injury [31] and hepatitis C virus infection [32] there are more than one hundred DE microRNAs or the ranges of DE microRNAs change can be up to tens or more than one hundred folds. Among the three lifestyles, CR had the mildest impact on microRNAs, DE microRNAs in EX changed to the biggest range.…”
