In the search of effective bioactive compounds, Co(II) (C1) and Cd(II) (C2) complexes of the type [M(FMAPIMP)(H2O)Cl].nH2O (where M = Co(II); n = 2, Cd(II); n = 3, and FMAPIMP = ligand[2‐((E)‐((2‐(((E)‐furan‐2‐ylmethylene)amino)phenyl)imino)methyl)phenol]) were synthesized and characterized using elemental analysis, UV–Vis., cyclic voltammetry, Fourier‐transform infrared (FT‐IR), nuclear magnetic resonance, and mass spectral studies. The thermal stability of nano‐sized Co(II) and Cd(II) complexes was studied using thermogravimetric analysis (TGA). Cobalt and cadmium oxides were synthesized using cobalt and cadmium nanoparticle (NP) structure Schiff base complexes as the raw material after calcination for 5 h at 600 °C. According to the results, Co(II) and Cd(II) complexes with mole ratio 1:1 of metal: H‐FMAPIMP which octahedral are the most probable geometry for it. On the contrary, synthesized C1 and C2 NPs were used as suitable precursors for the preparation of CoO and CdO NPs. The obtained NPs were characterized using FT‐IR, UV–Vis., TGA, powder X‐ray diffraction (PXRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and atomic force microscopy techniques. PXRD analysis revealed that the obtained oxides were crystalline and corresponded to CoO and CdO phases. Crystal size, shape, and morphology were determined using SEM and TEM. H‐FMAPIMP and its two complexes (C1 and C2) were tested against human ovarian cancer cell line (PA‐1). The synthesized Co(II) and Cd(II) complexes exhibited enhanced activity against the tested bacterial (Staphylococcus aureus and Escherichia Coli) and fungal strains (Candida albicans and Aspergillus fumigatus) as compared to H‐FMAPIMP. The results of the DNA‐cleavage activity indicated that the ligand and its two complexes can cleave calf thymus‐DNA at different degrees. Further, antituberculosis activity was performed using microplate alamarBlue assay. Among all these synthesized compounds, C1 exhibited good cleaving ability compared to the newly synthesized C2. Finally, the geometry of H‐FMAPIMP and its Co(II) and Cd(II) complexes was optimized using molecular modeling.

Porphyrin–Coumarin Dyads: Investigation of Photophysical Properties and DNA Interactions

Two new nonconjugated porphyrin-coumarin dyads with different orientations with respect to donor-acceptor entities and their zinc complexes were synthesized. Single-crystal structures of the free-base porphyrin-coumarin dyads were successfully resolved. The absorption spectra of the dyads were linear combinations of the spectra of their corresponding monomers, indicating a negligible electronic communication between the coumarin and porphyrin moieties. However, the fluorescence emission of the coumarin entity in all of the dyads was quenched significantly compared to that of pristine coumarin, and this effect was attributed to intramolecular energy transfer from the coumarin to the porphyrin. The energy transfer kinetics from the coumarin to the porphyrin was shown to be fast ( k = 1.13 × 10 s for the ortho-isomer and 5.13 × 10 s for the para-isomer in DMF) and efficient (transfer efficiency ca. 96-97%). Transient absorption studies showed that the excited state decay process (S → S*, S* → S, S → S, and S → T) of the para-isomer was faster than that of the ortho-isomer in DMF. All of the synthesized dyads were tested for their interactions with ct-DNA and photocleavage activity toward PBR322-DNA. The results revealed that all of the dyads interacted with ct-DNA via only an external groove-binding mode; the binding constants were calculated to be 3.24 × 10 (3a), 3.05 × 10 (3b), 3.04 × 10 (4a), and 4.88 × 10 (4b), and the photocleavage activity was in the order 4b < 3b < 4a < 3a. Furthermore, only the zinc complexes of the porphyrin-coumarin dyads could be absorbed by tumor cells (A549). These complexes were mainly localized in the cytoplasm, exhibited red fluorescence, and showed low cytotoxicity toward all of the tumor cell lines tested. The results showed that these zinc complexes of the porphyrin-coumarin dyads have potential applications in fluorescence imaging.

Novel cobalt, nickel and copper complexes were synthesized by regular reflux method and nano sized Co(II), Ni(II) and Cu(II) metal complexes were synthesized by a facile hydrothermal method using green solvent at various temperatures without the addition of any capping agent. The structural characterization was done by magnetic susceptibility, molar conductance, elemental analysis, thermal analysis, FT-IR, 1 H NMR, 13 C NMR, ESI mass, UV-Visible analysis. The morphology and size of the nano metal complexes were determined using FE-Scanning electron microscopy, powder X-ray diffraction data and atomic force microscopic techniques. All the spectral and analytical results reveal 1:2 metal to ligand ratios having [ML 2 (H 2 O) 2 ] stoichiometry, here M=Co(II), Ni (II) and Cu(II), L=deprotonated ligand. The non-electrolytic nature of the compound was confirmed by molar conductance experiment. The synthesized Schiff base and its metal complexes (7, 8 and 9) were tested for their biological activity. All the tested compounds exhibit decent anticancer and DNA cleavage activity and copper complex shows better activity results than other tested compounds.
