“…So far, bovine chimaeras have been obtained by aggregation of blastomeres from 2-to 8-cell embryos (Fehilly et al, 1984b), by aggregation of early morulae (Brem et al, 1984;Picard et al, 1986) and by injection of inner cell mass (ICM) cells into a blastocoelic cavity (Summers et al, 1983).…”

“…Sporadically, chimeras have been obtained from embryos of two species which do not normally interbreed. In domestic animals interspecific chimeras have been obtained from embryos of two subspecies of cattle, Bos taurus and B. indicus (Summer et al, 1983), and between the sheep and goat (Fehilly et al, 1984;Polzin et al, 1987;Roth et al, 1989;Ruffing et al, 1993). The karyotypes of B. indicus and B. taurus differ only in the morphology of the Y chromosome; the two species can normally mate with each other, and their offspring are fertile.…”

“…Using this technique at this stage of development, it appears that bringing half-or quarter-embryos together within one zona pellucida produces better results than does juxtaposing whole embryos (Brem et al, 1984;Summers et al, 1983;Picard et al, unpublished observations). The zonae pellucidae of two embryos are opened and a part (or all) of one is transferred to the zona containing a part (or all) of the other, using a pipette under the control of a micromanipulator.…”

“…However, ICMs can also be dissected manually (Fehilly et al, 1984a(Fehilly et al, , 1984bPicard et al, 1989). Similar injection of cells isolated from the ICM has been used to produce chimaeric laboratory animals (Gardner, 1968) and interspecific bovine chimaeras (Summers et al, 1983), but with a very low rate of success. The cells can be isolated from dissected ICMs after incubation in calcium-and magnesium-free culture medium containing 0.15% EDTA, 0.25% trypsin and 0.001% deoxyribonuclease I for 10-15 min.…”
