Thiazolidin-4-ones is an important heterocyclic ring system of a pharmacophore and a privileged scaffold in medicinal chemistry. This review is focused on the latest scientific reports regarding biological activities of thiazolidin-4-ones published in 2020 and 2021. The review covers recent information about antioxidant, anticancer, anti-inflammatory, analgesic, anticonvulsant, antidiabetic, antiparasitic, antimicrobial, antitubercular and antiviral properties of thiazolidin-4-ones. Additionally, the influence of different substituents in molecules on their biological activity was discussed in this paper. Thus, this study may help to optimize the structure of thiazolidin-4-one derivatives as more efficient drug agents. Presented information may be used as a practical hint for rational design of new small molecules with biological activity, especially among thiazolidin-4-ones.

“…Herein, it is interesting to note that the formation of AOX-Fe 2 + complex (ONN) as indicated in A, having relatively higher band gap energy (2.1 eV) as compared with B (N 2 O 2 ) AOX-Fe 2 + complex having the energy of 1.9 Ev. [37,38] From the obtained results, it is concluded that AOX acted as neutral tetradendate (N 2 O 2 ) donor to bind with Fe 2 + ion which was found to be more stable than the tridentate ONN donor binding of AOX to Fe 2 + ion. The frontier orbital gap results suggested that the complex with lower frontier orbital gap was more polarisable and reactive.…”

New coumarin‐oxazolyl derived receptor (E)‐3(2‐(2‐(1‐(2‐oxo‐2H‐chromen‐3‐yl)ethylidene)hydrazinyl)oxazol‐4‐yl)‐2H‐chromen‐2‐one (AOX) was designed, synthesised from the reaction of 3‐acetyl‐coumarin semicarbazone with 3‐bromoacetyl coumarin in chloroform/ethanol and the formation of AOX was confirmed by using different spectroscopic methods. The interaction of the AOX with different metal ions was investigated using photometric titrations and its selectively detected the Fe2+ ions.

Heterocyclic compounds containing nitrogen and sulfur, especially those in the thiazole family, have generated special interest in terms of their synthetic chemistry, which is attributable to their ubiquitous existence in pharmacologically dynamic natural products and also as overwhelmingly powerful agrochemicals and pharmaceuticals. The thiazolidin-2,4-dione (TZD) moiety plays a central role in the biological functioning of several essential molecules. The availability of substitutions at the third and fifth positions of the Thiazolidin-2,4-dione (TZD) scaffold makes it a highly utilized and versatile moiety that exhibits a wide range of biological activities. TZD analogues exhibit their hypoglycemic activity by improving insulin resistance through PPAR-γ receptor activation, their antimicrobial action by inhibiting cytoplasmic Mur ligases, and their antioxidant action by scavenging reactive oxygen species (ROS). In this manuscript, an effort has been made to review the research on TZD derivatives as potential antimicrobial, antioxidant, and antihyperglycemic agents from the period from 2010 to the present date, along with their molecular mechanisms and the information on patents granted to TZD analogues.
