Synthesis of bicyclic 1,4-thiazepines as novel anti-Trypanosoma brucei brucei agents

Abstract: Bicyclic thiazolidinyl-1,4-thiazepines were prepared and evaluated as potential antiparasitic agents. We found thiazepines with low micromolar EC50 against T. b. brucei and high selectivity index.

“…The novel compounds were tested for anthelmintic action against Nippostrongylus brasiliensis , Caenorhabditis elegans , and T. brucei brucei . When compared to the positive control (nifurtimox, IC 50 15.0 μM), two compounds ( 30 and 31) were found to be phenomenally effective, with IC 50 values of 2.5 μM and 2.8 μM against T. b. brucei (Vairoletti et al., 2019). Synthesis of some anti‐ T. cruzi agents of 5,7‐diaryl‐2,3‐dihydro‐1,4‐diazepines has been reported by Menezes et al.…”
