This work investigated the anti-amoebic activity of two samarium (Sm) complexes, the acyclic complex [bis(picrato)(pentaethylene glycol)samarium(III)] picrate-referred to as [Sm(Pic)(EO5)](Pic)-and the cyclic complex [bis(picrato)(18-crown-6)samarium(III)] picrate-referred to as [Sm(Pic)(18C6)](Pic). Both Sm complexes caused morphological transformation of the protozoa Acanthamoeba from its native trophozoite form carrying a spine-like structure called acanthopodia, to round-shaped cells with loss of the acanthopodia structure, a trademark response to environmental stress. Further investigation, however, revealed that the two forms of the Sm complexes exerted unique cytotoxicity characteristics. Firstly, the IC of the acyclic complex (0.7 μg/mL) was ~ 10-fold lower than IC of the cyclic Sm complex (6.5 μg/mL). Secondly, treatment of the Acanthamoeba with the acyclic complex caused apoptosis of the treated cells, while the treatment with the cyclic complex caused necrosis evident by the leakage of the cell membrane. Both treatments induced DNA damage in Acanthamoeba. Finally, a molecular docking simulation revealed the potential capability of the acyclic complex to form hydrogen bonds with profilin-a membrane protein present in eukaryotes, including Acanthamoeba, that plays important roles in the formation and degradation of actin cytoskeleton. Not found for the cyclic complex, such potential interactions could be the underlying reason, at least in part, for the much higher cytotoxicity of the acyclic complex and also possibly, for the observed differences in the cytotoxicity traits. Nonetheless, with IC values of < 10 μg/mL, both the acyclic and cyclic Sm complexes feature a promising potential as cytotoxic agents to fight amoebic infections.

Novel series of chrysin compound containing the organosulfur moiety from the Schiff bases derived from chrysin and 2-(phenylthio)aniline and their copper complexes were synthesized. They were characterized using analytical and spectral techniques. All the Cu(II) complexes exhibit the distortion square planar geometry. The in vitro antimicrobial activities of the investigated compounds were tested against the bacterial species, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Klebsiella pneumoniae and fungal species, Aspergillus niger, Fusarium solani, Rhizoctonia bataicola and Candida albicans by disc diffusion process. Free radical scavenging, superoxide dismutase, glutathione peroxidase and antioxidant activities of the copper complexes have also been studied. The DNA-cleavage activities of Cu(II) complexes, have been investigated by using agarose gel electrophoresis. Furthermore, interaction of copper complexes with bovine serum albumin was investigated by fluorescence spectrometry. The anticancer activities of the complexes have been carried out towards HeLa and HCT116 cancer cells. The pretreatment of chrysin compounds containing the organosulfur moiety of Cu(II) complexes ([CuHL4l(OAc)], [CuHL4m(OAc)] and [CuHL4n(OAc)]) elicited marked anti-inflammtory effects in carrageenan induced paw edema test in mice during various assessment times (1–5 hrs).

Bioactive complexes [La(L)(H 2 O) 3 Cl]Cl 2 .2H 2 O (1), [Er(L)(H 2 O) 3 Cl]Cl 2 (2), [Yb (L)(H 2 O) 2 Cl 2 ]Cl.4H 2 O (3), [Ru(L)(H 2 O) 2 Cl 2 ]Cl.H 2 O (4), and [Ta(L)(H 2 O)Cl 5 ](5) have been prepared from 2-acetylferrocene derivative Schiff base (L) and characterized with different spectroscopic tools. Infrared (IR) spectral data proved the action of the ligand as a neutral bidentate Schiff base, coordinating via N-azomethine and N-pyrazole. Ta(V)-complex was eight-coordinated complex, whereas others were six-coordinated complexes. Molar conductance results showed that La(III) and Er(III) chelates were 1:2 electrolytes, Yb(III) and Ru(III) chelates were 1:1 electrolytes, and Ta(V) chelate was non-electrolyte. Thermal behavior of all compounds was characterized by thermogravimetric analysis/differential thermogravimetric analysis (TG/DTG) curves with mass loss related to dehydration, coordination, and decomposition steps. Growth inhibitory effect and IC 50 values of compounds toward MCF-7 cancer cell line were measured, and the data revealed that all complexes showed higher anticancer activity than free ligand. The lowest IC 50 value was 4.5 μg/mL for La(III) and Ru(III) complexes. Furthermore, almost complexes showed good activities against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus species with inhibition zone diameter in the range of 10-15 mm/mg. While, they had no antifungal activities against Candida albicans and Aspergillus flavas species. Finally, the strong binding affinity of ligand and its complexes with different protein receptors was confirmed by MOE studies.
