Synthesis and evaluation of RGD peptidomimetics aimed at surface bioderivatization of polymer substrates

Abstract: AbstractÐSeveral RGD peptidomimetics have been prepared, in a convergent way, from the common ortho-aminotyrosine template (O-substituted with an anchorage-arm or a methyl group, and aN-substituted with a¯uorine tag for XPS analysis), and various o-aminoacid derivatives. The most¯exible compounds have shown a biological activity similar to that of the peptide reference (RGDS) in the platelet aggregation test. The compound 16a could be ®tted (by modelisation) with DMP 728 and c(RGDfV), two cyclic peptides that … Show more

“…Longer, more specific sequences have higher affinities for activated platelets than the GRGDS peptide. However, the length of the peptide can have dramatic effects on its temperature stability (26) as well as production costs. We designed these synthetic platelets to be stable at room temperature to facilitate their potential administration in the field.…”

“…Covalent immobilization of RGD peptides / small protein fragments on polymeric surfaces is another approach for surface modification of polymers [65,77]. Some of the studies that have applied this approach include covalent immobilization of RGD peptides on polystyrene surfaces with the help of polyethylene oxide [40], RGD peptidemodified poly(lactic acid-co-lysine) (PLAL) wherein, the RGD peptide was chemically cross-liked with lysine residue of PLAL with the help of 1,1-carbonyl diimidazole [78], and covalent immobilization of RGD peptides, collagen and fibronectin protein on polyvinyl alcohol surfaces [79][80][81].…”
