Synthesis and antitumor evaluation of some new derivatives and fused heterocyclic compounds derived from thieno[2,3‐b]pyridine: Part 2

Abstract: As a continuation of our research on developing anticancer agents and based on the proven proprieties of thieno[2,3-b]pyridines as anticancer, we have designed to synthesize novel thieno[2,3-b]pyridine derivatives that incorporate different biologically active heterocycles through various chemical reactions. All of the newly obtained compounds, compared with the standard anticancer drug (doxorubicin), were screened in vitro for their antitumor activity against hepatocellular carcinoma (HepG-2) and human breast … Show more

“…Heterocyclic compounds are widely used in agrochemicals, pharmaceuticals and veterinary products [5,6] . Biaryl skeletons are ubiquitous in many natural products and pharmaceutical molecules, including the indole biaryl scaffold, which has been shown to possess a variety of potent biological properties [7,8] . There are a variety of indole‐based natural compounds, including alkaloids, hormones, antibiotics, vitamins, amino acids, dyes, hemoglobin and pigments.…”

“…[5,6] Biaryl skeletons are ubiquitous in many natural products and pharmaceutical molecules, including the indole biaryl scaffold, which has been shown to possess a variety of potent biological properties. [7,8] There are a variety of indole-based natural compounds, including alkaloids, hormones, antibiotics, vitamins, amino acids, dyes, hemoglobin and pigments. A wide range of N-heterocyclic compounds, including anticancer agents, has been created by combining different heterocyclic rings.…”

“…[ 20 ] Polysubstituted pyridine containing cyano group exhibited antitumor activity in numerous publications. [ 21–27 ] In addition, cyano pyridine derivatives have shown to possess promising anti‐microbial, [ 28–30 ] antioxidant, [ 31–33 ] antibiotic, [ 34–36 ] anti‐inflammatory, [ 37,38 ] analgesic, [ 39 ] anti‐convulsant, [ 40 ] and anticancer activities. [ 41 ] Also, pyridine is widely distributed in many biological molecules, and found in drugs such as nifedipine.…”

“…What's more, further near-term literature investigation has revealed that adjunction of pyridine, halogen moiety, or peroxyalkyl can significantly increase the antitumor activity of candidate compounds. [26][27][28] Motivated by these findings, a series of novel 3-thioether-4-indolimino-4H-1,2,4-triazole derivatives bearing pyridyl moiety (17a~17r) were designed and synthesized by ethyl picolinate, ethyl isonicotinate, 1H-indole and indolin-2-one as raw materials. The anti-tumor activity against A549, PC-3, HepG2 and K562 cells was evaluated by methyl thiazolyl tetrazolium (MTT) assay.…”
