Synthesen in der Reihe der Indole und Indolalkaloide, VIII<sup>1)</sup> Darstellung und Drehungsverhalten von optisch aktivem 4a‐Methyl‐1.2.3.4‐tetrahydro‐4a<i>H</i>‐carbazol, seinem Methojodid und der zugehörigen Alkyliden‐indolin‐Base

Es wird gezeigt, daI3 eine aus 4a-Methyl-2,3,4,4a-tetrahydro-l-carbazolcarb~dehyd (5) dargestellte dimere Verbindung die Struktur 12 und nicht die friiher angenommene Struktur des offenen Aldehyds 7 bzw. 11 besitzt. Die Bildung des Racemats 12 auch beim Einsatz optisch aktiver Edukte sowie ein als Konkurrenzreaktion zur Dimerisierung ablaufender Halogen-Sauerstoff-Austausch zwischen dem vinylogen Imidoylchlorid 6 und der 1-Formyl-Verbindung 5 werden naher untersucht. Contributions to the Chemistry of Carbazol Derivatives with the Skeleton of Dmeric Calebash Curare Alkaloids, I Investigations an the Structure of a Model Compound for the Chromophore of the UltracurinesIt is shown that a dimeric compound synthesized from 4a-methyl-2,3,4,4a-tetrahydro-l-carbazolcarbaldehyde (5) as a model substance for the chromophore of the ultracurines has the structure 12 and not the structure 7 or 11 supposed earlier. The formation of the racemic compound 12 even from the optically active educts 6a and 5b as well as the halogen-oxygen exchange between 6 and 5, which competes with the dimerization, are investigated.Die Ultracurine A und B, zwei offensichtlich dimere Verbindungen bisher unbekannter Struktur, entstehen aus dem Calebassen-Curare-Hauptalkaloid C-Curarin-I (1) bei mehrstiindigem Erhitzen mit konzentrierter Salzsaure neben dem ebenfalls aus Curare isolierten Alkaloid C-Curarin-III(2) -3). Ultracurin-A 1aBt sich seinerseits durch Erhitzen mit 6 N HCI in Ultracurin-B und C-Curarin-I11 iiberfuhren ' ). Die Ultracurine besaI3en zum Zeitpunkt ihrer Isolierung (1958) eine besondere Bedeutung im Hinblick auf den damals noch nicht endgiiltig gesicherten zentralen Strukturbereich von C-'I H. Fritz und H. Die bereits in Erwagung gezogene Struktur 1 und die langwellige, dem UV-Spektrum von C-Curarin-111 ahnliche Absorption der Ultracurine gaben AnlaD zu der Vermutung, da13 Ultracurin-A die Partialstruktur 3 besitzen konnte ' ). Nachdem es gelungen war, rnit der aus 4a-Methyl-2,3;4,4a-tetrahydro-lH-carbazol (4) durch zweifache Vilsmeier-Reaktion (iiber das N-Formyl-Derivat) leicht zuganglichen Verbindung 5 einen Modellchromophor rnit nahezu der gleichen UV-Absorption wie C-Curarin-111 zu erhalten und damit dessen Struktur zu sichern lo), nahmen Fritz, Krekel und Meyerg) die Synthese der Verbindung 7 in Angriff, deren UV-Spektrum den vermuteten Chromophor 3 von Ultracurin-A bestatigen oder ausschlieBen sollte.Dazu wurde der Aldehyd 5 rnit Phosgen (oder besser mit P0Cl3 'I)) in das vinyloge Imidoylchlorid 6 iibergefuhrt, das in Ether bei Raumtemperatur rnit der aquimolaren Menge 5 umgesetzt wurde. Aus der Reaktionsmischung konnten zwei dimere Produkte isoliert werden: ein nicht naher untersuchtes ,,Indolin" (55 %, Schmp. 216"C, Zers.)...

in Wiley InterScience (www.interscience.wiley.com).Tetrahydrocarbazoles 4 obtained from phenylhydrazines and cyclohexanones gave by cyclocondensation with 2-substituted malonates 5 in all cases 4-hydroxy-8,9,10,11-tetrahydropyrido[3,2,1-jk]carbazol-6-ones 6 by attack at the nitrogen and the aromatic ring of tetrahydrocarbazoles 4; the direction of the cyclization was not dependent on substituents either in the aromatic or the saturated ring; isomeric pyridocarbazoles could not be isolated. Electrophilic substitutions of pyridocarbazoles 6 under mild conditions took place exclusively at the 5-position and gave pyridocarbazolediones 9-11 with 5nitro-, 5-hydroxy or 5-chloro-substituents. Exchange of the chloro substituent in 11 gave 5-azido-and 5-amino products 12, 14 or 16. Reactions at the aromatic ring were not observed. Chlorination of 4hydroxypyridocarbazoles 6 with phosphoryl chloride by nucleophilic substitution took place exclusively at the 4-position and gave 4-chloropyridocarbazolones 17, which were further reacted to azides and amines 18, 19.
