Synergistic inhibitory effects of cetuximab and curcumin on human cisplatin‑resistant oral cancer CAR cells through intrinsic apoptotic process

Abstract: Cetuximab, an epidermal growth factor receptor (EGFR)-targeting monoclonal antibody (mAb), is a novel targeted therapy for the treatment of patients with oral cancer. Cetuximab can be used in combination with chemotherapeutic agents to prolong the overall survival rates of patients with oral cancer. Curcumin is a traditional Chinese medicine, and it has been demonstrated to have growth-inhibiting effects on oral cancer cells. However, information regarding the combination of cetuximab and curcumin in drug-resi… Show more

“…Another possible synergistic interaction is with anti-EGFR therapy, targeting the EGFR/MAPK (mitogen-activated protein kinase) pathway known to stimulate cell proliferation [41]. Chen et al [42] observed that simultaneous treatment with the anti-EGFR antibody cetuximab and CUR demonstrated suppression of the EGFR/MAPK pathway in oral cancer cells resistant to platinum.…”

“…Cetuximab and the mixture of curcumin mediated apoptosis and signi icantly improved caspase-3 and caspase-9 behaviors relative to solitary therapy demonstrated oral anticancer results. The protein expression levels of Epidermal Growth Factor Receptor (EGFR) and Mitogen-activated protein kinase (MAPKs) were also signi icantly reduced by combination therapy (Chen, 2018).…”

“…Actually, an previous report has screened almost 600 herbal and natural compounds and found that CUR could promote EGFR degradation to potentiate the inhibitory effect of gefitinib on gefitinib-resistant lung cancer cells in vitro and in vivo [56]. Moreover, synergistic inhibitory effects of cetuximab and CUR on cisplatin-resistant oral cancer cells have also been documented recently [57]. Furthermore, after screening 36 CUR analogues, DMC was demonstrated to show the best inhibitory effects on both wild-type and mutant EGFR [58].…”
