Synergistic effect of nicorandil and amlodipine on mitochondrial function during isoproterenol-induced myocardial infarction in rats

Abstract: The synergistic effects of nicorandil (KATP-channel opener) and amlodipine (calcium-channel blocker) on heart mitochondrial enzymes and the mitochondrial antioxidant defence system was examined on isoproterenol-induced myocardial infarction in rats. The rats given isoproterenol (150 mg kg(-1) daily, i.p.) for two days showed significant changes in marker enzymes, mitochondrial enzymes and the mitochondrial defence system. Pre-co-treatment with nicorandil (2.5 mg kg(-1) daily, p.o.) and amlodipine (5.0 mg kg(-1… Show more

“…It has been reported that these enzymes are released from the heart into blood stream, thus increasing their concentration in serum (Wexler and Kittinger, 1963;Sathish et al, 2002;Deepa and Varalakshmi, 2003). In the present study, marked elevation in the activities of these enzymes in the serum of adriamycin intoxicated rats were observed.…”

“…A significant decrease in the activities of antiperoxidative enzymes, SOD and CAT may be due to diminished scavenging of HO • formed by ISO-induced lipid peroxidation. 37 A decrease in the activity of GPx makes mitochondria susceptible to ISO-induced myocardial damage that leads to mitochondrial dysfunction. 38 The decrease in the activities of GPx and GST may be due to a reduced availability of their substrate, GSH.…”

“…et al . [2627] Vascular reactivity to NA, Adr, AngII and 5-HT was measured by invasive blood pressure technique in all groups of rats, as described earlier by Balaraman et al . Endogenous enzymes such as catalase and superoxide dismustase are the first line cellular defense free radical scavenging enzymes against oxidative injury.…”
