Syndecan‐2 can promote clearance of T‐cell receptor/CD3 from the cell surface

Abstract: SummaryT cells express the heparan sulphate proteoglycans syndecan-2 and syndecan-4. Syndecan-4 plays a T-cell inhibitory role; however, the function of syndecan-2 is unknown. In an attempt to examine this function, syndecan-2 was expressed constitutively in Jurkat T cells. Interestingly, the expression of syndecan-2 decreased the surface levels of T-cell receptor (TCR)/ CD3 complex, concomitant with intracellular retention of CD3e and partial degradation of the TCR-ζ chain. Immunofluorescence microscopy revea… Show more

“…119,120 Syndecan-2 was found to be expressed in haematopoietic cells and myeloid cells, 121 and functionally reported to upregulate upon T cell activation and play a significant role in CD3 downregulation, through degradation of the T cell receptor (TCR). 122 These findings strongly suggest that enrichment of syndecan-2 expression in MSCs could play an essential role in immune modulation in injured tissue.…”

“…For example, CD73 positivity on MSC is associated with increased capacity for self-renewal and differentiation (36). The presence of Syndecan-2 (CD-362+) on MSC has been associated with enhanced immunomodulatory properties through downregulation of CD3+ cells by degradation of the T-cell receptor (37).…”

“…Syndecan family consists of four distinct members that are mostly expressed on epithelial, endothelial, neural, or fibroblastic cells, but they are also detected on haemopoietic cells (17,18). Syndecan 2 and syndecan 4 are up-regulated upon CD4+ T cell activation and act as inhibitors by promoting T cell receptor (TCR) clearance or by activating tyrosine phosphatase CD148 (19,20). Syndecan 1 (CD138) is commonly used as a marker to identify plasmablasts and plasma cells (21).…”
