Synchronized Orchestration of miR-99b and let-7g Positively Regulates Rotavirus Infection by Modulating Autophagy

Abstract: Rotavirus (RV), the major etiological agent of viral gastroenteritis in young children, kills over 200 thousand infants each year. In spite of available vaccines, rotaviral diarrhoea is still a major problem in developing countries of Asia and Africa. Therefore, the studies on RV infection and host antiviral responses are warranted. The active correlation between virus infection and activation of autophagy machinery and positive influence of autophagy on RV replication have been documented recently. Previous s… Show more

“…In contrast, functionality of miRNA is retained, as miRNA mimics successfully knocked‐down target expression both at early and late hours of RV‐SA11 infection (Figure e,f). This is in agreement with our previous findings, where miRNA mimics preserved their target repression potency in context of RV‐SA11 infection (Chanda, Nandi, & Chawla‐Sarkar, ; Mukhopadhyay et al, ). We also did not find global change of miRNA population characteristic of abrogated miRNA biogenesis in our miRNA microarray data (Chanda et al, ).…”

“…miR-99b modulates the expression of mTOR following RV infection and modulates autophagy for successful RV replication. 145 Molecular assays for detection and accurate quantitation of hepatitis C virus (HCV) RNA have been important for identification and management of the hepatitis C. ATG5, a target gene for miR-130a, significantly upregulated HCV replication and downregulated interferon-stimulated gene expression. And, miR-130a regulates host antiviral response and HCV replication through targeting ATG5 via the ATG5-dependent autophagy pathway.…”

“…Four miRNAs overexpressed in cataract patients were known to inhibit autophagy. 37 40 The miR-23b directly targeted the 3′-untranslated region of autophagy-related gene 12 mRNA to suppress the activation of neuronal autophagy. 41 A few miRNAs with decreased expression in cataract patients are known to promote autophagy.…”
