The productivity of pepper plants is strongly influenced by climatic and weather conditions as well as pests and diseases. One of the major losses in chili farming is the pest attack, such as whiteflies (Bemisia tabaci Genn.), a virus vector. Evaluation of the resistance of pepper plant germplasm against Begomovirus infection needs to be carried out, evaluate genetic diversity, estimate heritability values, and obtain a selection character for the resistance of chili plants Begomovirus. This research was conducted at Experimental Garden, Faculty of Agriculture, Universitas Syiah Kuala, and the Laboratory of Genetics and Plant Breeding, Faculty of Agriculture, Universitas Syiah Kuala from January to May 2020. Three varieties Gada, Anies and Selekta with seven test genotypes i.e F6 Gada, Seloka 4-10-2-2, F5136074-1-4-3, F5074136-2-3-2, F5074035-2-1-2, F5074035-2-4-2, F5074077-1-1-3 were screened for their resistance against Begomovirus. This study was designed using a non-factorial Randomized Block Design (RBD) and scoring parameter Begomovirus symptom. Tukey’s HSD, the standard deviation of genetic variance, heritability, and correlation were performed to analyze the data. The lowest disease intensity occurred in genotype F6 Gada with a percentage of 12.08% disease intensity (slightly resistant), and the highest level of disease intensity occurred in F5074035-2-4-2 with a percentage of 30.94% (susceptible). Variables symptom scoring, disease intensity, and incubation period were the most effective variables for selecting pepper resistance against Begomovirus infection based on broad-sense heritability values.

<p>Yellow leaf curl disease caused by Pepper yellow leaf curl virus (PepYLCV), member of geminiviruses group, is responsible for heavy yield losses for chili pepper production. Resistant genes which can cause immunity to the disease have not been found in germplasm collection. The aim of the research was to edit proliferating cell nuclear antigen (PCNA) gene by using CRISPR/Cas9 technology for developing plant resistance against geminivirus in chili pepper. A CRISPR/Cas9 plasmid cassette construct harboring the guide RNA of PCNA gene was constructed by Golden Gate cloning strategy. The construct was then introduced into chili genome via in planta method using Agrobacterium tumefaciens EHA105. The transformed plants were bioassayed by virus inoculation and confirmed using PCR and DNA sequencing to identify a mutagenesis event in PCNA gene target. The results showed that CRISPR/Cas9 plasmid cassette harboring gRNA of PCNA gene was successfully constructed. In planta transformation using A. tumefaciens vector harboring CRISPR/Cas9-gRNA PCNA construct resulted in 307 and 193 transformed plants from chili var. Lingga and Ciko, respectively. Bioassay by using virus inoculation to the transformed plants obtained 6 and 14 lines of Lingga and Ciko, respectively, which were resistant to geminivirus (no symptom observed). The resistant lines of chili pepper var. Lingga and Ciko were mutated in PCNA gene with one base insertion or deletion mutation types. These results exhibit that the CRISPR/Cas9 genome editing can be used to induce mutant of PCNA gene in chili pepper. Further investigation is necessary to evaluate the selected chili lines resistant to PepYLCV infection.</p>

Pepper yellow leaf curl virus (PYLCV) is a threat to chili plants and can significantly reduce yields. This study aimed as a pilot project to detect PYLCV by analyzing compounds emitted by chili plants using gas chromatography-mass spectrometry (GC-MS). The samples investigated in this research were PYLCV-infected and PYLCV-undetected chili plants taken from commercial chili fields. The infection status was validated by using a polymerase chain reaction (PCR) test. A headspace technique was used to extract the volatile organic compounds emitted by plants. The analysis of GC-MS results began with pre-processing, analyzing sample compound variability with a boxplot analysis, and sample classification by using a multivariate technique. Unsupervised multivariate technique principal component analysis (PCA) was performed to discover whether GC-MS could identify PYLCV-infected or not. The results showed that PYLCV-infected and PYLCV-undetected chili plants could be differentiated, with a total percent variance of the first three principal components reaching 91.32%, and successfully discriminated between PYLCV-infected and PYLCV-undetected chili plants. However, more comprehensive studies are needed to find the potential biomarkers of the infected plants.
