Meiotic cells of zebrafish have been prepared to show synaptonemal complexes (SCs) by light and electron microscopy. Completely paired SCs from both spermatocytes and oocytes were measured to produce an SC karyotype. The SC karyotype resembles the somatic karyotype of zebrafish and has no recognisable sex bivalent.Measurements of total SC length indicate that SCs grow longer and develop centromeres during pachytene. Oocytes consistently have longer SCs than spermatocytes, presumably correlated with the reported higher recombination frequency in females than in males.

SummarySynaptonemal complex (SC) complements prepared for electron microscopy allow high-resolution chromosome analysis. SC preparations were used to determine the relative lengths of the chromosomes of the Arabidopsis thaliana genome. This complements the extensive genetic and molecular analyses of these chromosomes. The two smallest SCs (chromosomes) carry a nucleolus-organizing region (NOR) at or near one end, and at prophase I of meiosis both have a relative SC length of about 16-17% of the total complement. The three longest SCs have relative lengths of 19,22.5 and 25% and can be identified on this criterion. Fluorescent in situ hybridization with nonradioactively labelled ribosomal DNA confirmed that there are two pairs of NOR chromosomes and also showed that the ribosomal genes are localized with the nucleolus. SC formation at NORs is often interrupted and this may reduce recombination in these regions of the genome.

The hermaphroditic flatworm Dendrocoelum lacteum shows a very pronounced sex difference in chiasma frequency; oocytes have 66 per cent more chiasmata than spermatocytes. The ultrastructural basis of this difference was investigated by three-dimensional reconstruction of synaptonemal complexes from ten oocytes and ten spermatocytes at pachytene and smaller numbers of zygotene nuclei. Oocyte nuclei at pachytene are larger and have longer synaptonemal complexes than the equivalent spermatocyte stage. The difference in synaptonemal complex length (60 per cent) corresponds very closely to the difference in chiasma frequency shown by oocytes and spermatocytes and suggests that synaptonemal complex (SC) length may contribute to the regulation of chiasma frequency. Recombination nodules were observed in both spermatocytes and oocytes but their quantitation was unreliable because they are small and differentiation from the unusually dense and prominent central region is difficult. Analysis of the few zygotene nuclei available shows that pairing is predominantly proterminal in both spermatocytes and oocytes, but oocytes may have several additional interstitial initiations. These extra initiations are likely to be necessary for efficient pairing of the longer oocyte chromosomes.
