Synaptonemal complex karyotype of zebrafish

Abstract: Meiotic cells of zebrafish have been prepared to show synaptonemal complexes (SCs) by light and electron microscopy. Completely paired SCs from both spermatocytes and oocytes were measured to produce an SC karyotype. The SC karyotype resembles the somatic karyotype of zebrafish and has no recognisable sex bivalent.Measurements of total SC length indicate that SCs grow longer and develop centromeres during pachytene. Oocytes consistently have longer SCs than spermatocytes, presumably correlated with the reporte… Show more

“…6A-7), unlike mouse and guinea pig pachytene spermatocytes in which g-H2AX signals are present but restricted to the XY chromatin. This observation is consistent with previous observations that no heteromorphic sex chromosomes exist in zebrafish (Traut and Winking, 2001;Wallace and Wallace, 2003). Hence, g-H2AX patterns are a powerful tool to characterize the timing and distribution of DSBs in zebrafish spermatogenesis.…”

“…6). By contrast, the gene family in zebrafish (Wallace and Wallace 2003), medaka (Ueda and Naoi 1999), and opossum (Rens et al 2001) is located near the centromere. To examine whether other genes present in subtelomeric regions of different chromosomes are similarly conserved across eutherian species, we carried out synteny analysis between human, mouse, and dog genomes using the Ensembl genome browser.…”

“…Since chromosome spreads with high resolution have already been reported in fish [46,47], we applied these methods to medaka spermatocytes. However, the resulting spreads did not react to the antibodies against MeSYCP1 and MeSYCP3, because of the loss of antigenicity due to strong spreading conditions during the sample preparation.…”
