Surface plasmon resonance imaging for affinity analysis of aptamer–protein interactions with PDMS microfluidic chips

Abstract: We report on the use of PDMS multichannels for affinity studies of DNA aptamer-human Immunoglobulin E (IgE) interactions by surface plasmon resonance imaging (SPRi). The sensing surface was prepared with thiol-terminated aptamers through a self-assembling process in the PDMS channels defined on a gold substrate. Cysteamine was codeposited with the thiol aptamers to promote proper spatial arrangement of the aptamers and thus maintain their optimal binding efficiencies. Four aptamers with different nucleic acid … Show more

“…Label-free and real-time analyses can be carried out with high throughput and low sample consumption by coupling microfluidic devices with the SPRI apparatus. [9,10] However, the use of SPRI for genomic assays is limited by A C H T U N G T R E N N U N G reduced sensitivity in the detection of hybridized DNA or RNA samples, so a number of different strategies directed towards amplifying SPRI response to DNA and RNA hybridization have recently been investigated, [11,12] in particular with use of colloidal gold nanoparticles (AuNps). [13][14][15] On the other hand, an-A C H T U N G T R E N N U N G other issue that deserves attention is the sequence specificity, in particular when single-nucleotide polymorphisms (SNPs) or point mutations are concerned.…”

“…Recent progress in this area as well as in data numerisation, transmission and processing allows now the construction of arrays of microorganisms or of enzymes arranged on a single detection platform for the determination of several parameters at the same time. In parallel, the development of new biomimetic receptors such as that molecularly imprinted polymers (MIP) or aptamers (synthetic oligonucleotides) is expanding to overcome the fragility of natural bioreceptors (Wang et al, 2007;Guan et al, 2008). Methods allowing a more efficient immobilisation of the bioreceptor will also have to be developed to improve the robustness and sensitivity of biosensors.…”
