Surface Antigens on Eosinophils in Bronchial Asthma and Eosinophilic Pneumonia

Abstract: Surface antigens on peripheral blood eosinophils from 23 patients with bronchial asthma, 6 with eosinophilic pneumonia and 8 controls were examined using a new direct method. Peripheral blood eosinophils in bronchial asthma and eosinophilic pneumonia showed higher complexity and/or granularity than those from controls. The percentage expression of HLA DR, CD4 and CD45RO on peripheral blood eosinophils from patients with bronchial asthma were increased compared with those from patients with eosinophilic pneumon… Show more

“…Eosinophils in the sputum of asthmatics have been shown to express HLA-DR (Hansel et al 1991). Airway, but not blood, eosinophils in chronic eosinophilic pneumonia also have been demonstrated to express HLA-DR (Beninati et al 1993, Okubo et al 1995, Sakamoto et al 1995b). In patients with asthma, even blood eosinophils have been found to express greater HLA-DR than eosinophils from normals (Sakamoto et al 1995a).…”

Moreover, comparisons of blood and bronchoalveolar lavage eosinophils obtained 48 hr after segmental antigen (Sedgwick et al 1992) or 4-6 hr after inhalational (Mengelers et al 1994) challenges in allergic subjects have demonstrated that HLA-DR was expressed on airway eosinophils.…”

“…Based on the pattern of forward and side scatter, a granulocyte gate was set. Neutrophils were identified as CD9-negative granulocytes (Saito et al, 1986;Sakamoto et al, 1995). Data on CD9-positive eosinophils are not presented due to the low number of cells.…”

“…After lysis o f red cells, peripheral blood leukocytes were washed thoroughly with phosphatebuffered saline and analyzed by flow cytometry' with FACScan (Bccton Dickinson) according to a new method described previously [3]. Briefly, eosinophils were identified by positivity for CD9 (Nichirei, Tokyo) in the granulocyte fraction gated by autofluorescence (eosino phils were more than 99% in purity), and were analyzed for expres sion of HLA-DR, CD4, C D llb , CD44 and CD45RO (Nichirei, Tokyo).…”

“…Recent studies have suggested that blood eosinophils from normal control subjects and from patients with eosinophilia vary with respect to their chemotactic re sponses [1], cytotoxic ability [2] and surface antigens [3]. This heterogeneity of eosinophils may be associated with differences in inflammatory environment or in cytokine expression patterns.…”
