Supramolecular Polymerization‐Induced Nanoassemblies for Self‐Augmented Cascade Chemotherapy and Chemodynamic Therapy of Tumor

Abstract: The clinical application of chemodynamic therapyis impeded by the insufficient intracellular H 2 O 2 level in tumor tissues.H erein, we developed as upramolecular nanoparticle via asimple one-step supramolecular polymerization-induced self-assembly process using platinum (IV) complex-modified b-cyclodextrin-ferrocene conjugates as supramolecular monomers.T he supramolecular nanoparticles could dissociate rapidly upon exposure to endogenous H 2 O 2 in the tumor and release hydroxyl radicals as well as platinum … Show more

“…Versatile CDs were synthesized according to our previous study, 33 of which playing the functions of multi‐imaging and PTT. 1,1′‐Ferrocenedicarboxylic acid was introduced not only as the “glue” for crosslinking but also as an effective CDT agent 31,34 . First, the successful fabrication of Fc‐CD NPs was investigated via transmission electron microscopy (TEM) measurements.…”

“…10 ). 56 Specifically, the supramolecular NP can dissociate rapidly when reacting with H 2 O 2 in TME and then in situ produce ˙OH as well as platinum( iv ) drug, similarly, to overcome the limitation of conventional CDT, platinum( iv ) drug could be reduced into cisplatin to activate NOX for enhancing H 2 O 2 level. Additionally, the size of the supramolecular nanoparticle is tunable and could be readily excreted from the body via renal clearance, thus both enhancing the EPR effect and minimizing the systemic toxicity and side effects.…”

“…Imaging probes, therapeutic agents and targeting ligands can be integrated into the theranostic system through self-assembly on the basis of supramolecular chemistry, greatly avoiding tedious organic synthesis [ 5 7 ]. Additionally, the dynamic and reversible nature of non-covalent interactions endow the supramolecular theranostics with brilliant stimuli-responsiveness, activating the loaded drugs in sites of action triggered by the specific tumor microenvironment, possibly optimizing the theranostic performance and decreasing undesirable side effects [ 8 10 ]. Various non-covalent interactions have been employed to construct supramolecular theranostics, such as H-bonds, van der Waals forces, metal coordinations, electrostatic interactions and host–guest complexations [ 11 , 12 ].…”
