Supplementation with a complex of active nutrients improved dermal and epidermal characteristics in skin equivalents generated from fibroblasts from young or aged donors

Abstract: Cultured skin equivalent (SE, Mimeskin) was generated by co-culturing skin fibroblasts and keratinocytes on a collagen-glycosaminoglycan-chitosan dermal substrate. In order to examine donor age effect, fibroblasts from 19- (young) or 49- (aged) year-old females were used. Culture medium was supplemented with nutrients complex containing soy extract, tomato extract, grape seed extract, white tea extract, sodium ascorbate, tocopherol acetate, zinc gluconate and BioMarine complex. Epidermal and dermal structure a… Show more

“…Another cause of this significant difference on the Ki67 might be due to the excessive expression of Ki67 in the suprabasal layers of the native epidermis. Despite the fact that the Ki67 is mostly expressed in the basal layer of epidermis, a large number of Ki67‐stained suprabasal layers were observed in this study. We suspect that the frozen skin samples embedded in OCT compound were not sectioned in a vertical plane.…”

“…In addition, the free amino acids that constitute the 'natural moisturizing factor' involved in keeping the skin hydrated have their origin in the cleavage of profilaggrin into filaggrin [19,20] . However, our model does not allow studying skin permeability as described by Henning et al [21] , Mitriaikina et al [22] and Nicoli et al [23] , but testing activity and the tolerance of actives [24,25] .…”

“…Effectively, as multipotent stem cells, ASCs have the capacity to differentiate into fibroblasts [49]. Hence, in SE with 25% ASCs, the fibroblast-rich dermis provides a better quality DE, which in turn affects the quality of the epidermis, as previously shown [84,85]. Moreover, transdifferentiation of ASC into epithelial cells [50,[85][86][87][88][89][90] and migration toward the epidermis cannot be excluded.…”
