Superoxide anion production and some sperm-specific enzyme activities in infertile men

Abstract: The relationship between superoxide anion production and the activities of sperm specific NADH-diaphorase and isozyme LDH-C, has been investigated in 25 normozoospermic and 17 oligozoospermic semen samples. The samples were separated into high and low density sperm populations on discontinuous, two step (40%:80%) Percoll gradients prior to analysis. Superoxide anion generation was significantly higher (P< 0.001) and diaphorase activity significantly increased (P<0.01) in both low and high density sperm fractio… Show more

“…Our findings have shown that superoxide anion levels were higher in infertile men with abnormal semen parameters compared with normal group, and this is in accordance with the results of other studies (Gavella et al, 1995;Said et al, 2004). In asthenooligoteratozoospermic patients, the level of superoxide anion was the highest compared with other infertile groups.…”

“…It was previously established that after TritonX-100 treatment, both the soluble and membrane-bound diaphorase were measured (Gavella et al, 1995). For lactate dehydrogenase-C4 isoenzyme and NADH diaphorase activity determination, spermatozoa were permeabilized with 0.2% TritonX-100 (Sigma, St. Louis, MO, USA).…”

It was previously established that after TritonX-100 treatment, both the soluble and membrane-bound diaphorase were measured (Gavella et al, 1995). TritonX-100 treatment was also employed for LDH-C4 isoenzyme activity determination, as proposed by Casano et al (1991).…”
