Extraction of rutin from Ruta graveolens L. leaves was performed in choline chloride based deep eutectic solvents (DESs). Firstly, rutin extraction was performed in five different DESs; pure DESs and with different amounts of water added (25% and 50% v/v), for different times (1 h, 3 h and 6 h) in order to find the best DES for further optimization. Choline chloride plus citric acid (1 : 1) was shown to be best DES and optimization of rutin extraction was performed using response surface methodology (RSM) based on the selected solvent and applying different extraction parameters: extraction time, temperature and water content.The rutin content was determined by HPLC and varied from 0.55 g/100 g of plant to 1.81 g/100 g of plant depending on the applied conditions. The best conditions for rutin extraction were found to be: a temperature of 70°C, an extraction time of 52 min and a water content of 27 % for the extraction of 1.88 g /100 g plant.

Six medicinal plants Helichrysum italicum (Roth) G. Don, Angelica archangelica L., Lavandula officinalis L., Salvia officinalis L., Melilotus officinalis L., and Ruta graveolens L. were used. The aim of the study was to compare their extracts obtained by Soxhlet (hexane) extraction, maceration with ethanol (EtOH), and supercritical CO2 extraction (SC-CO2) targeted on coumarin content (by high performance liquid chromatography with ultraviolet detection, HPLC-UV), 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging capacity, and total phenols (TPs) content (by Folin–Ciocalteu assay). The highest extraction yields were obtained by EtOH, followed by hexane and SC-CO2. The highest coumarin content (316.37 mg/100 g) was found in M. officinalis EtOH extracts, but its SC-CO2 extraction yield was very low for further investigation. Coumarin was also found in SC-CO2 extracts of S. officinalis, R. graveolens, A. archangelica, and L. officinalis. EtOH extracts of all plants exhibited the highest DPPH scavenging capacity. SC-CO2 extracts exhibited antiradical capacity similar to hexane extracts, while S. officinalis SC-CO2 extracts were the most potent (95.7%). EtOH extracts contained the most TPs (up to 132.1 mg gallic acid equivalents (GAE)/g from H. italicum) in comparison to hexane or SC-CO2 extracts. TPs content was highly correlated to the DPPH scavenging capacity of the extracts. The results indicate that for comprehensive screening of different medicinal plants, various extraction techniques should be used in order to get a better insight into their components content or antiradical capacity.

Scopoletin (7-hydroxy-6-methoxy coumarin) is a phenolic coumarin isolated from many plants, known as an important compound of the phytoalexin group. This article is created to provide information regarding the synthesis and pharmacological activity of scopoletin. Scopoletin has been found in many plant species and isolated from various parts of the plant (roots, fruits, leaves, stems, etc.) such as Morinda citrifolia, Aegle marmelos, Erycibe obtusifolia Benth, Lasianthus lucidus Blume, Melia azedarach L., Sinomonium acutum, Convolvulus prostratus, and Solanum lyratum, etc. which all possess a number of medicinal properties. Its various pharmacological activities have been reported through a number of investigations. It is reported that such compounds produced specific biological activities and possible health implications for humans in food and medicine. Pharmacological activities that are established in-vivo are antithyroid, antihypertensive, anti-proliferative, anti-inflammatory, neurological, anti-dopaminergic and anti-adrenergic, antidiabetic drug, and antihyperuricemic activities. Based on in-vitro studies, scopoletin has pharmacological activities, including an antihepatotoxicity, antibacterial, antifungal, antitubercular, and antioxidant. From the assorted pharmacological activities of scopoletin, it has the potential to be further developed.INTRODUCTION: Scopoletin (7-hydroxy-6methoxy coumarin) is a phenolic coumarin isolated from many plants, known as an important compound of the phytoalexin group. It has a yellow crystalline structure with a molecular weight of 192 and a melting point of 204-206 °C. Its various biological activities have been reported through a number of investigations. Booth et al., (2004) reported that such compounds produced specific biological activities and possible health implications for humans in food and medicine.
