Sugar Functionalized Synergistic Dendrimers for Biocompatible Delivery of Nucleic Acid Therapeutics

Abstract: Sugars containing cationic polymers are potential carriers for in vitro and in vivo nucleic acid delivery. Monosaccharides such as glucose and galactose have been chemically conjugated to various materials of synergistic poly-lysine dendrimer systems for efficient and biocompatible delivery of short interfering RNA (siRNA). The synergistic dendrimers, which contain lipid conjugated glucose terminalized lysine dendrimers, have significantly lower adverse impact on cells while maintaining efficient cellular entr… Show more

“…Dendrimers consist of three main parts: a central core, a growing branch unit (called generation (G)), and a large number of terminal functional groups (usually amino groups) on the surface . The terminal groups of the dendrimer can be lipid-modified to obtain the dendrimer-lipids. In 2012, Yu et al synthesized a class of dendrimer-lipids with different alkyl chain lengths and dendrimer generations through click chemistry . The long alkyl chain of 389 (Figure ) promotes the assembly of siRNA/carrier, can form a stable self-assembled complex with siRNA, and increases the stability of the complex through hydrophobic interactions.…”

“…Considering that many substances of interest exhibit limited solubility in water, several attempts have been proposed to encapsulate such compounds, deliver them effectively, and release in a controlled manner at the site of action. Colloidal particles such as dendrimers, micelles, capsules, liposomes and polymersomes exhibit adequate size, high loading capacity, and sufficient stability which make them frequently used in the mentioned applications [ 10 , 11 , 12 , 13 , 14 , 15 , 16 , 17 ]. However, several aspects need to be considered during the design and development of such carriers.…”

“…It might be a technique for effective and biocompatible siRNA delivery. 83 Maltose-changed PPI dendrimers (44) are also glyco-nanoparticles as a result of their unmodied congeners dramatic increase in biocompatibility. However, as demonstrated by the discovery of PPI-m G4 dendrimers inside the fully modied dense shell variation stimulate the NF-kB pathway in a cell version of monocytesthe traditional effectors of innate immunity (Fig.…”
