Substituent effects in the <sup>13</sup>C NMR spectra of 17α‐ethinyl‐19‐nortestosterone derivatives

The structural effects of C-3 sustituents on two derivates of cholic acid have been studied by 1D-as well as homonuclear and heteronuclear 2D-NMR spectra. The stereochemical structure of ring A is derived from homonuclear vicinal coupling constants. The consequences of C-3 substitution on ' H and 13C chemical shifts, on stereochemical and electronical effects are discussed.Gallensauresalze sind wirksame Detergenzien, da sie sowohl polare als auch unpolare Regionen im Molekul besitzen. Sie sind Hauptbestandteil der Gallenflussigkeit und dienen als Losungsvermittler bei der Verdauung von Lipiden. Durch Micellenbildung wird die Oberflache der Lipide vergrohert und dadurch die Hydrolyse der Lipide durch Lipasen gefordert und ihre Resorption im Dunndarm erleichtert *I.Da die Hydroxygruppe in der C-3-Position essentiell ist, stellt sich die Frage, ob die Konfiguration an C-3 fur die physiologischen Eigenschaften von Bedeutung ist. Aus diesem Grund wurden im folgenden die Strukturparameter von 3a,7a,12a-Triacetoxy-5P-cholsaure-methylester (1) und 3P-Methoxy-7a,12a-diacetoxy-5P-cholsauremethylester (2) als Derivate der Cholsaure und deren Konfigurationsisomeren an C-3 untersucht. Die Konfigurationsbestimmung erfolgt mit Hilfe der NMR-Spektroskopie. Gerade bei Steroiden liefert diese analytische Methode rasche und detaillierte Strukturinformation 3-6).Im Gegensatz zum Cholsaurederivat 1, dessen Stereochemie und '3C-Spektren7) bekannt sind, war von der synthetischen Verbindung 2 zwar die cis-Verknupfung der Ringe A und B nach der Darstellung naheliegend, aber die Konfiguration an C-3 und die Konformation des Ringes A (Sessel-oder Wannenform) unklar.
