Study on Chemical Constituents in Polygoni Cuspidati Folium and its Preparation by UPLC-ESI-Q-TOF-MS/MS

Abstract: A rapid and credible analytical method was developed using online UPLC-ESI-Q-TOF-MS/MS to identify chemical constituents in Polygoni cuspidati folium and its preparation. By accurate mass measurements within 6.5 ppm error for [M-H]- ion in routine analysis, 26 chemical constituents, including tannin, derivatives of phenylpropionic acid, stilbene, flavonoid, anthraquinone, torachryson and its derivatives, were identified or tentatively characterized. Among them, five constituents (compounds 19-23) were firstly … Show more

“…The precursor ion of compound 39 is m/z 193.0506 [M − H] − , and fragment ions are m/z 178.06 [M − CH 3 ] − , m/z 149.0611 [M − H − CO 2 ] − , and m/z 134.0375 [M − H − CO 2 − CH 3 ] − were obtained, which is consistent with the fragmentation pattern of ferulic acid reported in the literature, 32 so it is determined to be ferulic acid.…”

“…Both of the deprotonated molecular ions yield [M − H + CH 3 COOH − H 2 O] − at m / z 503 and m / z 665. Compound 39 produced their [M − H + CH 3 COOH − 2H 2 O] − , [M − H − CH 2 ] − , [M − H − Glc − CO] − , [M − H − Glc − C 5 H 10 O 5 ] − and [M − H − Glc − C 5 H 10 O 5 − CH 2 ] − ions at m / z 647, m / z 609, m / z 433, m / z 311 and m / z 297 [52]. Compound 46 produced their [M − H + CH 3 COOH − CO 2 ] − , [M − H − C 4 H 8 O 4 ] − , [M − H − C 5 H 10 O 5 − CH 2 ] − and [M − H − C 5 H 10 O 5 − C 9 H 8 O 2 ] − ions at m / z 477, m / z 341, m / z 297 and m / z 163.…”
