Study of uroproteins in myeloma patients by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE)

Abstract: Samples of the urine of 10 myeloma patients with proteinuria were examined by SDS-PAGE. Light chain proteins of Bence Jones (B.J.) type were excreted by 7 patients as monomer (m.w. 20--26.5 x 10(3) Dalton, by 2 patients as a mixture of monomer and dimer and by one patient as dimer. By two-dimensional electrophoresis in SDS-PAG and subsequent electrophoresis in agarose containing kappa and lambda chain specific antibodies the immunological identity of monomeric and dimeric B.J. protein of one patient has been s… Show more

“…1,2 SDS-AGE and SDS-PAGE, electrophoretic techniques that separate proteins according to molecular mass, have demonstrated FLCs in the 20-26.5 kDa region. 3,4 The relationship between the identification of urinary FLCs and the development of renal disorders has been established in multiple myeloma, where renal amyloidosis (AL type), light-chain deposition disease, and myeloma cast nephropathy represent different renal manifestations of the same disease. 18 In dogs, FLCs have been observed in urine from 25 to 45% of patients with multiple myeloma or chronic lymphocytic leukemia.…”

“…2,3 Sodium dodecyl sulfate electrophoresis in humans identifies FLCs with a molecular mass of 20-26.5 kDa. 3,4 In dogs, identification of urine FLCs generally is carried out with the heat test, 5-7 but immunoprecipitation 7,8 and SDS-PAGE 9 also have been used.…”

“…In two patients (both k), the proportion of LC D to LC M increased over a four-month period of medication (cyclophosphamide) possibly suggesting ªescapeº of the neoplastic clone from treatment [87]. SDS-PAGE (first dimension) in combination with agarose gel crossed immunoelectrophoresis (second dimension) was subsequently used to investigate BJP (n = 10) to identify the free LC and determine its molecular weight (M r 20 000±26 500) and size distribution (seven patients showed LC M only; 2, LC M + LC D; and 1, LC D only) [88]. In a methodological development, SDS-PAGE of BJP (n = 12) was demonstrated on horizontal ultrathin 4±25% polyacrylamide gradient gels using the Multiphor electrophoresis system (Amersham Pharmacia Biotech UK, Little Chalfont, Bucks, UK) ± the free LC and LC fragment (M r 15 000) were detected by silver staining following electrophoresis of unconcentrated urine and were disElectrophoresis 1999, 20, 1307±1324 Electrophoresis of BJP 1311 …”
