The sugar composition of the surface of isolated human keratinocytes has been investigated using lectins. A method is described in which the FITC-labelled lectin is displaced with competing sugar prior to measurement. This avoids errors due to aspecific binding and utilizes the minimum biological sample. Evidence is presented which indicates that little cell surface glycoconjugate is lost during the preparation of the keratinocyte suspension from human epidermal slices using the technique described here. Values for keratinocytes are similar to those reported for fibroblast lines, being much higher than those found using erythrocytes or lymphocytes.

Psoriatic hyperproliferative epidermis is characterized by a decreased beta 2-adrenergic adenylate cyclase response as well as by altered differentiation markers that include decreased loricrin and increased involucrin. Using a quantitative reverse transcription-polymerase chain reaction, we analysed the expression of beta 2-adrenergic receptor-mRNA, loricrin-mRNA, and involucrin-mRNA in the epidermis of five patients with psoriasis vulgaris. The mRNAs of the beta 2-adrenergic receptor and loricrin in the involved epidermis were significantly decreased, by 0.35-fold (P < 0.01) and 0.55-fold (P < 0.05) respectively, compared with uninvolved epidermis. In contrast, the involucrin mRNA expression of the involved epidermis was significantly increased, by 3.77-fold (P < 0.01). No significant difference in beta-actin mRNA transcripts was detected between the involved and the uninvolved epidermis. These results indicate that the altered expression of the beta 2-adrenergic receptor, loricrin, and involucrin, in the psoriatic involved epidermis, is associated with different amounts of each mRNA transcripts.

Large melanosome complexes in the human gingival epithelium

The morphology of melanosome complexes (MC) in gingival epithelium from smokers and non‐smokers was studied by electron microscopy. The cross‐cut diameter was measured on 770 complexes within keratinocytes. In pigmented smokers and non‐smokers the average diameter was between 0.51 and 0.63 μm, which is larger than those observed in clinically non‐pigmented non‐smokers and compared to those previously reported from clinically non‐pigmented Caucasians. The diameter of 95% of all complexes was less than 1.0 μm, but 40 large melanosome complexes (LMC) were also found. LMC could reach a diameter of 3.2 μm, and contain more than 100 melanin granules. The presence of LMC in oral mucosa has not been reported previously, but they have been observed in pigmented human skin disorders. Our finding of LMC in the gingival epithelium of pigmented individuals among smokers and non‐smokers, but not in non‐pigmented non‐smokers, indicate that this entity is related to melanosome packaging in situations of high melanin production and not necessarily only to pathological conditions.
