Background: Polyscias fruticosa is used widely as food, remedy for diseases, and as an ornamental across Afro-Asian countries. For instance, P. fruticosa is used traditionally as an anti-asthma, anti-tussive, and a muco-suppressant herbal remedy for asthmatics in Ghana. Although many studies have investigated the pharmacological basis of the ethnobotanical uses of P. fruticosa, however, its effect on the reproductive system remains completely unknown. Aim of study: This study assessed effects of P. fruticosa leaf extract (PFE) on male fertility and toxicity in adult male Wistar rats. Materials and methods: After crude preparation of PFE, it was subjected to qualitative phytochemical, thin layer chromatography and gas chromatography mass spectrometry analyses. The effect of PFE was assessed on male fertility and toxicity by using healthy adult male Wistar rats. Rats were randomly assigned to normal saline (5 ml/kg po, n = 5), Clomiphene Citrate (CL; 50 mg/kg po; n = 5), and PFE (100, 200, and 500 mg/kg po; n = 5, respectively) groups and treated for 21 days. On day 22, rats were sacrificed and male fertility parameters (left testis weight, relative testis weight, caudal epididymal weight, caudal epididymal sperm count, sperm motility, sperm morphology, and assessment of male sex hormones and testicular histology) were assessed. Results: There were no significant changes in bodyweight, weight of left testis, weights of right and left caudal epididymides between treatment groups (PFE and CL) and control. Caudal epididymal sperm count increased in PFE (100 and 500 mg/kg)-treated rats relative to control. Sperm motility relatively increased in PFE-treated rats compared to control. Sperm abnormality decreased in PFE-treated rats, especially in PFE (100 mg/kg) group compared to control. Serum testosterone levels decreased inversely with serum luteinizing hormone levels in PFE-treated rats compared to control. There were minimal-to-no-alterations in histological sections of testis, except vacuolations at primary spermatocyte stage. Glycosides, saponins, cyanogenic glycosides, sterols, and alkaloids were detected in PFE. Conclusion: PFE improved caudal epididymal sperm count and may be useful as male fertility enhancer but exhaustive safety studies on key male sex organs needs to be established.

BackgroundEvaluation of Euphorbia dracunculoides of family Euphorbiaceae during previous studies had established the in vitro antioxidant and in vivo anti-inflammatory activities. The plant is used by the local communities of Pakistan for various disorders including rheumatism and edema. In this investigation we have evaluated the hepatoprotective effects against CCl4 induced toxicity in rat.MethodsDry powder of the aerial parts of E. dracunculoides was extracted with 95% methanol to get the extract (EDME). To investigate the hepatoprotective effects of EDME the Sprague-Dawley male rats were divided in to 8 groups with 6 rats in each. Group I and II were the normal and vehicle treated while the Groups III-VI were injected intraperitoneally with 1 ml of CCl4 (30% in olive oil). Rats of Group IV were orally administered with silymarin (50 mg/kg) while the Group V and VI with 200 mg/kg and 400 mg/kg of EDME, respectively. Animals of Group VII (200 mg/kg) and VIII (400 mg/kg) were treated with EDME alone. The treatments were given thrice a week for 4 weeks. Effects of EDME were evaluated for the protective effects against oxidative stress and genotoxicity induced with CCl4 in liver of rat.ResultsAnalysis of serum indicated significant (p < 0.05) rise in the level of aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and globulin whereas decrease was recorded for the total protein and albumin in CCl4 treated rats. In liver tissues the activity level of catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), glutathione-S-transferase (GST), glutathione (GSH) was decreased while the level of lipid peroxides; thiobarbituric acid reactant substances (TBARS), nitrite and hydrogen peroxide increased in CCl4 treated rats as compared to the control group. Histopathological injuries and DNA damages were recorded in liver of rat with CCl4 treatment. However, co-administration of EDME, dose dependently, ameliorated the CCl4-induced hepatic toxicity in these parameters.ConclusionsThese results suggested that the phyto-constituents of EDME were able to ameliorate the oxidative stress induced with CCl4 and can be a useful therapeutic agent for oxidative stress related disorders.

BackgroundIn the present study the antioxidant potential of a methanol extract of Phyllanthus emblica leaves (PELE) was determined by in vitro methods as well as by an in vivo animal model, along with HPLC-DAD screening for phyto-constituents.MethodsThe in vitro antioxidant potential of PELE was assessed by scavenging of DPPH, nitric oxide and anti-lipid peroxidation assays. For in vivo evaluation, a 60-day experimental plan was followed in which Sprague Dawley rats were administered with 1 mL/kg of CCl4 (CCl4 : DMSO + Olive oil; 30 % v/v) alone or with different doses of PELE (200, 400 mg/kg p.o.). Silymarin (100 mg/kg) as standard drug was also administered to CCl4 treated rats. HPLC-DAD analysis was performed to quantify polyphenolic phytochemicals.ResultsPELE exhibited an appreciable in vitro antioxidant activity and scavenged the DPPH radical (IC50 = 39.73 ± 2.12 μg/mL) and nitric oxide (IC50 = 39.14 ± 2.31 μg/mL) while for anti-lipid peroxidation moderate antioxidant activity was noticed. Reduced levels of antioxidant enzyme activities viz., superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and reduced glutathione (GSH) whereas enhanced levels of total extractable proteins, lipid peroxides (TBARS), nitrite and H2O2 were induced by CCl4 administration in lungs of rat. Co-administration of PELE to rats exhibited a dose dependent decline in the oxidative injuries induced in these parameters. Histopathological damages such as disrupted alveoli, infiltration of macrophages and modified architecture of Clara cells was reversed to the normal state with co-administration of PELE. HPLC-DAD analysis indicated the presence of gallic acid, rutin, kaempferol and caffeic acid in the PELE.ConclusionThe findings of this study demonstrate that presence of polyphenolics and other active constituents in PELE might play a significant role in repairing the pulmonary damages instigated with CCl4.
