Studies on New Activities of Enantiomers of 2-(2-Hydroxypropanamido) Benzoic Acid: Antiplatelet Aggregation and Antithrombosis

Abstract: R-/S-2-(2-Hydroxypropanamido) benzoic acid (R-/S-HPABA), a marine-derived anti-inflammatory drug, however, the antiplatelet and antithrombotic effects have not been investigated. In this paper, the in vitro antiplatelet activities and in vivo antithrombotic effects of R-/S-HPABA were investigated, for the first time. The effects of R-/S-HPABA on platelet aggregation induced by adenosine diphosphate (ADP), collagen (COLL) and arachidonic acid (AA) were evaluated. In addition, the in vivo bleeding time, clotting… Show more

“…The marine yeast-derived R-/S-2-(2-Hydroxypropanamido) benzoic acid (R-/S-HPABA) and the extracts of the alga Eisenia bicyclis were shown to reduce thrombus weight in rats and/or mice and exert a strong inhibition of platelet aggregation in vitro [58,59]. These effects appear to be mediated through distinct mechanisms: R-/S-HPABA likely inhibits cyclooxygenase-1 (COX-1) activity and thromboxane B2 (TXB2) formation, while Eisenia extracts reduce P2Y 12 downstream signalling, a process crucial in platelet activation and aggregation [58,59].…”

“…Significantly, R-/S-HPABA inhibited the thromboxane B2 level and elevated F1α generation. In addition, R-/S-HPABA could lower the weight of carotid thrombosis ( 35 ).…”

“…R -/ S -2-(2-Hydroxypropanamido) benzoic acid ( R -/ S -HPABA) is a small organic molecule ( Figure 5 D) with anti-inflammatory and analgesic properties [ 40 ]. It was first isolated from the fermentation broth of marine fungus Penicillium chrysogenum .…”

“…Moreover, this benzoic acid derivative has COX 1 inhibitory potential, which seems to be stereoselective, since the S isomer was found to be more efficient than the R isomer. The study proposed that the antithrombotic activity of the molecule was somewhat correlated with the inhibition of COX 1 activity [ 40 ].…”
