Diabetes mellitus is a metabolic syndrome considered one of the life-threatening diseases in the last two decades. This research aimed to investigate the anti-diabetic potential of bitter honey collected from Nilgiris using both in vitro and in vivo methods. The mineral content of bitter honey was also estimated using atomic absorption spectrophotometer. Bitter honey had a higher amount of zinc and copper, while heavy metals like lead, nickel, and cadmium were below the detection limit. The in vitro antidiabetic study was performed using alpha-amylase and alpha-glucosidase inhibition methods. Acute toxicity (OECD 423) was conducted in female Wistar rats to determine the lethal dose of bitter honey. The antidiabetic activity was carried out in type-2 diabetic Wistar Albino rats induced with streptozotocin and nicotinamide. The experimental rats were categorized into five groups (n=8): the normal group, the diabetic control group, standard glibenclamide-treated diabetic group, bitter honey 200 mg/kg, and 400 mg/kg b.w. treated diabetic group. After the treatment period (28 days), blood samples were collected for biochemical studies, and the pancreas was dissected for histopathological studies. The in vitro antidiabetic studies revealed the antidiabetic potential of bitter honey compared to standard acarbose. Treatment of diabetic rats with bitter honey revealed a statistically significant reduction (P<0.05) in the levels of fasting blood glucose (FBG) compared to untreated diabetic rats. This was accompanied by an elevated HDL and a decrease in LDL, VLDL, triglycerides, total cholesterol, SGOT, SGPT, urea, and creatinine. Histopathological changes in the pancreas indicated a marked improvement in a dose-dependent manner. The study concluded that bitter honey could potentially decrease the levels of FBG in diabetic rats and the various biochemical and histopathological abnormalities associated with diabetes mellitus.

Abutilon indicum (L.) is one of the traditional medicinal plants and its extract has been utilized for antidiabetic activity. Diabetes mellitus is a metabolic disorder characterized by hyperglycemia and its occurrence is increasing fast in most countries. Peroxisome proliferator-activated receptor gamma (PPARγ) plays an important role in adipogenesis. The present study determines the effect of A. indicum methanolic leaf extract as potential antidiabetic inhibitors. The in vitro analysis was carried out by using 3T3L1 for glucose uptake assay, RIN5F cell lines for insulin secretion, hydrolysis assay to predict glycemic index, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging assay, and molecular interaction of the known bioactive compounds with PPARγ. The experimental results showed 100 nM insulin and 50 mM LiCl glucose uptake with 3.37 and 3.86 fold stimulation, respectively, when compared to the control, whereas the given samples of 200, 400, and 800 µg/ml showed 1.37, 1.62, and 1.85 µg/ml fold stimulation, respectively, when compared to the control. The insulin release in RIN-5f cells was observed with a positive control and crude extract and the results showed 4.67 and 2.67 µg/ml, respectively. The hydrolysis index value was found to be 53.30 and the glycemic load was 17.48 µg/ml. In the DPPH assay, the sample showed dose-dependent DPPH radical scavenging activity with an IC 50 value of 99.12 µg/ml when compared to standard quercetin with an IC 50 value of 1.7 µg/ml. The molecular interaction of PPARγ and active methyl trans-p-coumarate (−5.44) > methyl caffeate (−4.49) > syringic acid (−3.9) > pinellic acid (−2.62) compared with thiazolidinediones (−7.62) formed a novel type of oral antidiabetic medication that improved metabolic management in type 2 diabetic patients by increasing insulin sensitivity. The overall result shows that A. indicum (L.) is a potential indicator for sensitizing insulin secretion and strongly inhibits the release of glucagon which can be used as a therapeutic agent for treating and managing diabetes.

Objective: In this study, the leaves of medicinal plant Bauhinia racemosa Lam. with different pharmacological activities were subjected to phytochemical screening and assessment of their in vitro inhibitory potential with porcine pancreatic α-amylase enzyme to treat and management of diabetes.Methods: Plant leaves were extracted sequentially with ethanol solvent. A modified 3,5-dinitrosalicylic acid method was adopted to screen α-amylase inhibition assay. The ethanol extract was analyzed qualitatively and gas chromatography-mass spectrometry analysis technique for the active phytoconstituents according to the standard protocols.Results: A phytochemical screening of leaves extract reveals the presence of carbohydrate, alkaloids, saponin, glycosides, steroids, tannins, flavonoids, triterpenoid, and phenolic compounds. The ethanol extract reported inhibition of α-amylase enzyme activity at IC 50 value 61.72 ± 0.03 µg/mL and acarbose as a standard drug at IC 50 value 28.07 ± 0.02 µg/mL. Conclusion:The results of the study indicate that B. racemosa Lam. leaves contain some of bioactive phytochemicals might to be exhibiting in vitro antidiabetic activity, which was leading to decreases the rate of starch digestion.
