Abstract:In Malaysia black pod disease caused by Phytophthora palmivora is one of the serious problems causing yield loss in cocoa plantation. In this study, an endophytic bacterium living within tissues of healthy Theobroma cacao L. leaves was isolated and assessed its ability to inhibit the growth of P. palmivora. Based on molecular identification using 16S rDNA gene sequences analysis, this endophytic bacterium was identified as Bacillus amyloliquefaciens which was subsequently named as Bacillus amyloliquefaciens LKM-UL. Preliminary test using dual culture method showed B. amyloliquefaciens LKM-UL has strong ability to inhibit the growth of P. palmivora. B. amyloliquefaciens showed the highest antifungal activity after 24 h incubation in nutrient broth. Cells-free supernatant from B. amyloliquefaciens showed the highest antifungal activity of 30.6 mm with cell concentration of Log 10 10.63 CFU/mL via agar disc diffusion method. B. amyloliquefaciens LKM-UL also produced plant growth hormone identified as 1-naphthalene acetic acid (NAA), tryptamine, 3-indole propionic acid (IPA), indole-3-butyric acid (IBA), gibberellic acid (GA), trans-zeatin and kinetin.

Abstract:The future need for sustainable agriculture will be met in part by wider use of biological control of plant pathogens over conventional fungicides hazardous to the environment and to public health. Control strategies involving both (i) direct use of microorganisms antagonistic to the phytopathogen, and (ii) use of bioactive compounds (secondary metabolites/antibiotic compounds) from microorganisms on the phytopathogen were both adapted in order to investigate the ability of streptomycetes isolated from the rhizosphere of strawberry plants to promote the growth of the fruit and suppress Botrytis cinerea causing strawberry rot on the Sunshine Coast, Queensland, Australia. In vitro studies showed that 25/39 streptomycetes isolated from strawberry field soils inhibited B. cinerea growth by antifungal activity, ranging from antibiosis to volatile compound production. However, when non-volatile antifungal compounds were extracted and applied aerially to the actively growing strawberry fruits infected with B. cinerea, a significant disease reduction was not recorded. On the other hand, plant and fruit growth was promoted by the presence of actively growing streptomycetes in container media. Findings might indicate that live streptomycete inoculum can be used as growth promoting agent in container media for this economically important crop.

Wilt desease of banana caused by Fusarium oxysporum f.sp. cubense (FOC) is one of the most destructive deseases of banana in the tropics. Actinomycetes are the most economically and biotechnologically valuable prokaryotes able to produce wide range of bioactive secondary metabolites. The aims of the present study are to isolate and screen the actinomycetes with high potential ability to produce secondary metabolites that have inhibitory activity against plant pathogenic fungi, Fusarium oxysporum f. sp. cubense. Two isolates from Lampung and Cianjur showed activity against fungi. The isolates designed as L.3.1 and CiIA5b. The metabolites from potent stain was produced by extraction of culture filtrate with ethyl acetate : methanol (4 : 1), it was tested for their antifungal activity by well diffusion method. Evidence for in vitro antibiosis of L.3.1 and CiIA5b isolates was demonstrated by the zone of fungal-growth inhibition. Production of secondary metabolites was analysis by thin layer chromatography (TLC) and bioautography assays. In this study, the metabolites from L.3.1 and CiIA5b have showed good antifungal activity.
