Studies of the humoral factors produced by layered chondrocyte sheets

Abstract: The authors aimed to repair and regenerate articular cartilage with layered chondrocyte sheets, produced using temperature-responsive culture dishes. The purpose of this study was to investigate the humoral factors produced by layered chondrocyte sheets. Articular chondrocytes and synovial cells were harvested during total knee arthroplasty. After co-culture, the samples were divided into three groups: a monolayer, 7 day culture sheet group (group M); a triple-layered, 7 day culture sheet group (group L); and … Show more

“…Hamahashi et al reported that a layered chondrocyte sheet produced the most humoral factors, including PGE2, which plays a key role in its anti-inflammatory function. 153 In chondrocyte sheets, catabolic factors such as MMP3, MMP13, and a disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) were also observed to decrease, while the expression of TIMP1 with antagonistic actions against MMP3 increased.…”

“…21 In addition, Hamahashi et al showed that layered chondrocyte sheets produced higher levels of TGFb and PGE2 as humoral factors compared with monolayer chondrocyte sheets and suspected that these factors might contribute to cartilage repair. 22 However, these previously reported results might not…”

“…We found that the formation of a chondrocyte sheet structure enhanced transforming growth factor-β secretion from the cells [37], which implies that maintaining the membranous structure after cryopreservation is a prerequisite for function. Therefore, in the present study, we focused on both the maintenance of sheet structures and of cell viability after vitrification.…”
